摘 要:利用响应曲面法(RSM)对鹿血多肽的制备工艺进行优化。在单因素实验的基础上,选取实验因素与水平,根据中心组合(Box-Behnken)实验设计原理采用3因素3水平的响应曲面法对各个因素的显著性和交互作用进行分析。结果表明:鹿血多肽制备的最佳工艺条件为酶解时间3 h,蛋白酶B的用量4%,酶解温度60℃,在此条件下,理论上酶解液中低聚肽所占比例为96.12%,提取率达100%。The preparation process of polypeptides from deer blood was optimized by response surface methodology in this study. The one-factor-at-a-time experiments were carried out to figure out the effects of four parameters on the preparation of polypeptides from deer blood. According to these results, the relationship of the ratio of oligopeptides in the hydrolysate with the other three parameters was mathematically modeled on the basis of a three-variable, three-level central composite design based on Box-Behnken model with 17 runs. And then the model established was subjected to analysis of variance to test the significance of various variables and response surface analysis to understand crresponding interactive effects. From the above investigations and analyses, optimum conditions were determined to be that hydrolysis time of 3 h, amount of protease B of 4% and temperature of 60 ℃. Under the optimum conditions, from the view of theoretical calculation, ratio of oligopeptides in the hydrolysate and extraction ratio of oligopeptides were 96.12% and 100%, respectively.
