摘 要:以柠檬酸钾(K_3C_6H_5O_7·H_2O)、柠檬酸三铵[C_6H_5O_7(NH_4)_3]和硫酸铵[(NH_4)_2SO_4]3种不同的盐为盐析剂,利用冻融破壁的方法提取巢湖新鲜蓝藻的藻蓝蛋白破壁液,研究3种不同的盐析剂对纯化藻蓝蛋白的影响。借用光谱扫描法分析判断不同盐纯化效果的优劣。实验结果表明:(NH_4)_2SO_4是纯化蓝藻中藻蓝蛋白的最适宜的盐析剂。在室温25℃下,一步盐析时(NH_4)_2SO_4的最佳浓度为1.2 mol/L,二步盐析时(NH_4)_2SO_4的最佳浓度为2 mol/L,所得藻蓝蛋白的纯度可达1.95,得率为1.71%。Three different kinds of salts of K_3C_6H_5O_7·H_2O C6H5O7(NH4)3 and (NH4)2SO4 were used as salting-out agents, and extraction in phycocyanin of fresh blue algae from Chaohu was obtained by employing method of freezing and melting to break cellwall. Then researches have been made on effects of purified cyanophycin that influenced by three different salting-out agents and judging the effects of salting-out by using spectral scanning method. The outcomes are as follows: (NH4)2SO4 is a more suitable agent for salting out phycocyanin from blue algae. At 25 % of indoor temperature, the best concentration of (NH4)2SO4 was 1.2 mol/L in step one and 2 mol/L in step two. The purity of phycocyanin could be 1.95 and the yield was 1.71%.
