摘 要:目的:观察激素性股骨头坏死兔模型骨组织中相关因子的表达情况,探讨激素性股骨头坏死的可能作用机制。方法:健康普通级新西兰家兔20只按完全随机原则分成两大组(试验组10只,对照组10只),试验组注射甲基泼尼松龙造激素性股骨头坏死兔模型。两组兔行X线片、MRI检查,并采用免疫组织化学检测股骨头标本骨组织中TGF-β1、MMP-2、BGP表达水平,采用qPCR检测股骨头标本骨组织中RunX2的表达水平。结果:试验组兔X线片检查、MRI检查以及病理检查均示股骨头坏死,对照组兔X线片检查、MRI检查以及病理检查均无股骨头坏死征象。免疫组织化学检测试验组兔股骨头骨组织中TGF-β1、MMP-2、BGP的表达结果较对照组低,差异均具有统计学意义(P<0.001);qPCR检测试验组兔股骨头骨组织中RunX2信号的表达结果较对照组低,差异均具有统计学意义(P<0.001)。结论:激素性股骨头坏死的机制可能与股骨头骨组织中的TGF-β1、MMP-2、RunX2、BGP表达水平过低相关。Objective: To observe the expression of bone related proteins in bone tissue of rabbit femoral head treated with glucocorticoid and explore its action mechanism in bone repair of glucocorticoid-induced necrosis of femoral head. Methods: 20 healthy and ordinary New Zealand rabbits were divided into two groups(10 for experiment group and 10 for control group) according to completely randomized principle. Ear intravenous injection of methylprednisolone has performed to the rabbits in experimental group to establish the glucocorticoid-induced necrosis of femoral head model. After the model was established, X radiographic examination and MRI examination had performed to rabbits of both two groups. After the image examination, pathological examination of specimens should be performed and test the expression level of TGF-β1, MMP-2, and bone gla protein in femoral head bone tissue of both two groups' rabbit with immunohistochemistry and test the expression level of RunX2 signal in femoral head bone tissue of rabbit with qPCR examination at last. Results: According to the X radiographic examination, MRI examination and pathological examination, experiment group showed femoral head necrosis, but the control group didn't. The expression of TGF-β1, MMP-2 and BGP in femoral head bone tissue from experiment group were all less than those from control group showed by immunohistochemistry, and the differences was statistical significance(P〈0.05). The expression level of RunX2 in femoral head bone tissue from experiment group were all less than those from control group showed by qPCR, and the difference was of statistical significance(P〈0.05). Conclusion: The mechanism of glucocorticoid-induced necrosis of femoral head might relate to the excessively low expression level of TGF-β1, MMP-2, BGP and RunX2 in femoral head bone tissue.
