摘 要:目的通过观察糖酵解通路中相关蛋白M2-型丙酮酸激酶2(PKM2)、哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)、磷脂酰肌醇3激酶(PI3K)、蛋白激酶B(AKT)、缺氧诱导因子1α(HIF-1α)在胃癌患者标本中的表达,以研究PKM2、mTOR、PI3K、AKT、HIF-1α等糖酵解通路中相关蛋白与胃癌之间的相关性。方法采用免疫组织化学法检测89例胃癌患者标本与20例活检正常的胃部组织标本中PKM2、mTOR、PI3K、AKT、HIF-1α等糖酵解通路相关蛋白的表达。结果胃癌组患者组织中PKM2、mTOR的平均光密度值依次为4 423.18±328.77;4 586.08±284.80,均显著高于对照组的相应指标(135.84±15.36,813.55±61.26),差异有统计学意义(P<0.05)。而PI3K、AKT、HIF-1α在二组中差异不明显,无统计学意义(P>0.05)。胃癌患者根据性别、年龄、分期、有无淋巴结转移、分化程度等临床特征进行分组,结果显示胃癌患者在不同性别、年龄及分化程度下PKM2和mTOR的表达率差异无统计学意义(P>0.05);在不同分期、淋巴结转移下PKM2和mTOR的表达率有差异,且具有统计学意义(P<0.05)。结论国人胃癌组织中PKM2和mTOR的相对表达量均显著高于非肿瘤人群,且其表达与胃癌的转移相关,提示PKM2和mTOR与胃癌之间具有相关性。Objective To observe the glycolytic pathway associated protein PKM2,mTOR,PI3 K,AKT,HIF-1αexpression in patients with gastric cancer specimens,to study the correlation between PKM2,mTOR,PI3 K,AKT,HIF-1αand other glycolytic pathway gastric cancer-associated protein.Methods Immunohistochemistry was used to detect the expression of 89 cases of patients with gastric cancer and 20 cases of biopsy specimens of normal gastric tissue specimens PKM2,mTOR,PI3 K,AKT,HIF-1αand other glycolytic pathway proteins.Results PKM2 gastric carcinoma patients,the mean optical density value of mTOR were 4 423.18 ± 328.77,4 586.08 ± 284.80,respectively,significantly higher than the corresponding index(135.84±15.36,813.55 ± 61.26)(P〈0.05),there were statistical significance.The PI3 K,AKT,HIF-1αwere not significantly different in the two groups,and no statistically significant(P〉0.05).Gastric cancer patients according to sex,age,stage,lymph node metastasis,differentiation and other clinical features were grouped in patients with gastric cancer showed varying degrees of sex,age and differentiation PKM2 expression of mTOR and no statistically significant(P〉0.05);in a different stage,lymph node under PKM2 expression and mTOR were different,the difference was statistically significant(P〈0.05).Conclusion Chinese gastric cancer PKM2 and the relative expression levels of mTOR are significantly higher than non-tumor population,and its expression and metastasis of gastric cancer,suggesting a correlation between PKM2 and mTOR in gastric cancer.
