摘 要:目的研究酶法提取龙眼核多糖的最佳工艺条件。方法在单因素试验的基础上,根据响应面法的Box-Behnke试验设计原理,采用加权评分法以龙眼核提取物中多糖得率和多糖含量为综合评价指标,建立二次多项式回归方程的预测模型,构建以综合评分为响应值的响应曲面和等高线图,考察酶解温度、酶解时间和pH值3个主要因素对多糖提取效果的影响以及因素间的交互作用。结果回归模型拟合性良好。各因素对总多糖提取效果的影响程度为:pH>酶解温度>酶解时间。最佳提取工艺条件为:料液比1∶30,pH 6.08,酶解温度54.47℃,酶解时间3.03 h,酶添加量为1.00%。结论酶法可提高龙眼核多糖的提取效率,且提取条件温和、污染小,优选的工艺稳定可行,可为相关生产加工企业提供有益的参考。OBJECTIVE To study the optimal technological conditions for extracting polysaccharides from longan seeds by composite enzyme method. METHODS On the basis of single-factor experiment and Box-Behnke design principle, the quadratic polynomial mathematical model, response surface and contour line map were analyzed. The effects of enzymatic hydrolysis time, temperature and pH value on the polysaccharides yield were explored by using yield and content of polysaccharides as evaluating indicators. RESULTS The degree of fitting and confidence level for mathematical model was satisfactory. The effect order of 3 factors on polysaccharides yield was as follows: pH value 〉 enzyme hydrolysis temperature 〉 enzyme hydrolysis time. Optimum extraction conditions were concluded as follows: liquid/solid ratio of 1:30, pH of 6.08, enzymatic hydrolysis temperature of 54.47 ℃, time of 3.03 h and pectase amount of 1.00%. CONCLUSION Enzymatic extraction was mild and highly efficient with little pollution. The optimal extraction process is steady and reliable, which could provide references for industrial production.
