摘 要:目的 探讨雷公藤内酯醇对大鼠大脑皮质内注射β-淀粉样蛋白(Aβ)后树突棘和发育调节脑蛋白(drebrin)表达的影响。方法 30只SD雄性大鼠随机分成对照组、Aβ组、用药组(每组10只)。Aβ组大鼠双侧大脑皮质各一次性注射凝聚态Aβ_(1~40),对照组大鼠大脑皮质注射等量生理盐水,用药组大鼠在大脑皮质注射Aβ_(1~40)后每日腹腔注射雷公藤内酯醇(0.4 mg/kg),共30 d。Golgi染色和免疫组织化学染色检测各组大鼠大脑皮质神经元树突棘的密度和drebrin的表达。结果 Golgi染色显示,Aβ组大鼠大脑皮质神经元树突棘密度明显低于对照组(P〈0.01),用药组大鼠大脑皮质神经元树突棘密度明显高于Aβ组(P〈0.05)。免疫组织化学染色显示,Aβ组大鼠大脑皮质drebrin免疫反应阳性颗粒平均光密度较对照组明显减少(P〈0.01);用药组大鼠大脑皮质drebrin免疫反应阳性颗粒平均光密度较Aβ组明显增加(P〈0.01)。结论 雷公藤内酯醇能抑制Aβ所致drebrin的丢失,减轻Aβ所致树突棘的损伤。Objective To investigate the effects of triptolide on dendritic spines and the expression of drebrin after microinjection of beta-amyloid protein(Aβ) into cerebral cortex in rats. Methods Thirty male SD rats were randomly divided into control,Aβ and triptolide- treated groups( n= 10). The Aβ group was given an bilateral injection of aggregated Aβ1-40in cerebral cortex in rats. The control group was given an injection of sterile saline in cerebral cortex. The triptolide-treated group was administered intraperitoneally triptolide (0.4 mg/kg) once daily :for 30 days after Aβ -4o injection. Spine density of cerebral cortical neurons was assayed by Golgi staining and drebrin expression of cerebral cortical neurons was assayed by immunohistochemical staining. Results The spine density of cerebral cortical neurons in Aβ group was obviously decreased compared with that 'of control group( P〈0.01 ) ;spine density of cerebral cortical neurons in triptolide-treated group was obviously increased compared with that of Aβ group(P〈0.05 ). The average optical density of drebrin immunoreactive product in cerebral cortex of Aβ group was obviously decreased compared with that of control group( P〈0.01 ) ;the average optical density of drebrin im- munoreactive product in cerebral Cortex of triptolide-treated group was obviously increased compared with that of Aβ group(P〈0.01 ). Con- clusions Triptolide could inhibit the loss of drebrin induced by Aβand alleviate the degeneration of dendritic spines induced by Aβ.
