摘  要:目的观察硫化氢(H2S)通过氧化应激及细胞因子调控四氯化碳(CCl4)诱导小鼠肝纤维化的抑制作用。方法将20只雄性昆明小鼠随机分为4组,正常对照组、肝纤维化组、低剂量Na HS组和高剂量Na HS组,每组5只。采用化学发光试剂盒检测氧化应激酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)水平;ELISA检测炎性细胞因子TNF-α、IL-1α和IFN-γ水平;全自动生化检测仪检测各组小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)和甘油三酯(TG)水平;Realtime-PCR检测参与氧化应激基因的表达调控。结果与正常对照组比较,肝纤维化组的SOD、CAT和GSHPx水平降低,细胞因子TNF-α、IL-1α和血清中ALT、AST、TG升高(P<0.05或0.01);与肝纤维化组比较,Na HS高、低剂量组的ALT、AST降低,SOD、CAT和GSH-Px升高,且呈剂量依赖关系(P<0.05或0.01),TG变化则无统计学意义(P>0.05)。结论 H2S可通过降低氧化应激水平,抑制细胞因子的释放和肝纤维化,改善肝功能。Objective To observe the inhibitory effect of hydrogen sulfide(H2S) regulating carbon tetrachloride(CCl4) to induce hepatic fibrosis through oxidative stress and cytokines. Methods 24 male Kunming mice were randomly divided into 4 groups, Normal Control Group, Hepatic Fibrosis Group, Low Dose Na HS Group and High Dose Na HS Group, 5 mice in each group. The level of SOD, CAT and GSH-Px was detected with chemiluminescence kit, and the level of inflammatory cytokines TNF-α, IL-1αand IFN-γ was detected with ELISA kits, AST, ALT and TG in mice serum was detected with automatic biochemical detector, and oxidative stress relative genes were detected with Real time-PCR. Results Compared with Control Group, the level of SOD, CAT and GSH-Px in Hepatic Fibrosis Group was decreased, while TNF-α and IL-1α and serum ALT, AST and TG in Hepatic Fibrosis Group were increased(P〈0.05 or 0.01). Compared with Hepatic Fibrosis Group, AST and ALT in High and Low Dose Na HS Group was decreased while SOD, CAT and GSH-Px was increased, which showed dosedependency(P〈0.05 or 0.01). There was no significant change in TG(P〉0.05). Conclusion H2 S can inhibit the hepatic fibrosis and improve liver function though inhibiting the release of cytokines and reducing the oxidative stress.
