摘 要:目的研究没食子儿茶素没食子酸酯(EGCG)对百草枯导致急性肺损伤保护作用的机制。方法选用60只清洁级Ⅱ级健康SD雄性大鼠分别建立对照组、百草枯组(PQ组)和PQ+EGCG干预组(PQ+EGCG组),每组各20只。24小时后分别取各组大鼠的肺泡灌洗液、肺组织进行肺组织干湿重比、肺泡灌洗液中mtDNA、TNF-α和IL-6测定以及肺组织HE染色。结果百草枯组肺泡灌洗液中mtDNA、TNF-α和IL-6明显升高,而经过EGCG预处理后,3项指标均明显下降(均P<0.05)。肺组织干湿重比和病理评分在百草枯组明显升高,说明肺组织出现明显炎性浸润;而在PQ+EGCG组中均明显降低(均P<0.05)。结论本研究结果发现,EGCG可以通过保护线粒体、减少mtDNA释放,降低百草枯导致的肺组织炎症反应水平,同时为百草枯导致急性肺损伤的机制研究和治疗提出了一个新的理论基础。Objective To study the mechanism of the protective effects of epigallocatechin-3-gallate(EGCG)on paraquat-induced acute lung injury.Methods 60 SD rats were divided into the control group,paraquat(PQ)group and PQ+EGCG group.After 24 h,broncho-alveolar lavage fluid(BALF)and lung tissue were collected to measure levels of mtDNA,TNF-αand IL-6,and wet/dry ratio and HE staining,respectively.Results Our data showed that levels of mtDNA,TNF-αand IL-6elevated in PQ group,which could be inhibited by EGCG(P0.05).Wet/dry ratio and pathological score of PQ group were higher than that of the control group,which could be also inhibited by EGCG(P0.05).Conclusion We find that EGCG can protect paraquat-induced acute lung injury through inhibiting the release of mtDNA.Meanwhile,we also provide a novel theory for understanding and treating paraquat-induced acute lung injury.
