摘  要:目的观察小鼠高脂饮食后S6K1 mRNA和蛋白表达变化,探讨S6K1在胰岛素抵抗发生中的作用。方法 40只C57BL/6小鼠随机分为正常饮食组(N)和高脂饮食组(H),8周后建立胰岛素抵抗模型。用ELISA法检测空腹血清中胰岛素值和小鼠OGGT曲线变化;采用Northern blot法检测骨骼肌中S6K1mNRA的表达;采用Western blot法检测骨骼肌中S6K1蛋白的表达。结果小鼠经过14周高脂饮食后;体重和血清中空腹胰岛素水平也都显著升高;且OGTT曲线显示糖耐量明显受损,已建立成胰岛素抵抗模型,发现S6K1蛋白和mRNA表达也显著升高。结论 S6K1在高脂饮食诱导胰岛素抵抗中发挥重要作用。Objective To study the role of S6K1 in high fat diet induced insulin resistance C57BL/6mice,we surveyed mRNA and protein expression of S6K1 in skeletal muscle of insulin resistance C57BL/6mice.Methods 8-month-old C57BL/6 male mice are divided into two groups:that is normal chow group(N)and high-fat diet group(H).The serum insulin level was measured by ELISA.Measured and recorded the curves of the OGTT.Test the level of S6K1 mRNA in skeletal muscle by Northern blot and S6K1 protein in skeletal muscle by western blot.Result In the study,mice fed for 14 weeks had found that the body weight and fasting serum insulin level of mice from high fat diet group were increased.Furthermore,glucose tolerance was impaired.So that we have modeled insulin resistance.At the same time,we discovered that the expression of S6K1 mRNA and protein were significantly increased in HC group comparing with NC group.Conclusion S6K1 plays an important role in the development of diet induced insulin resistance.
