干扰素γ基因多态性与动脉粥样硬化斑块稳定性相关性研究

摘　　要：目的初步探讨干扰素γ(interferon gamma,IFNγ)+2109A/G和+874A/T位点基因多态性是否影响动脉粥样硬化(atherosclerosis,AS)的发生及其斑块稳定性。方法选择204例住院患者,将AS斑块不稳定患者109例为观察组和无斑块或动脉粥样硬化斑块稳定患者95例为对照组,流式细胞仪检测血浆IFNγ水平,并通过PCR产物直接测序法检测IFNγ+2109基因表达,通过序列特异性引物引导的PCR反应技术检测IFNγ+874基因表达,采用DNAStar、GeneTool软件分析测序结果。结果观察组IFNγ+2109位点AA、AG、GG型基因为46.79%、44.04%、9.17%,对照组分别为43.16%、43.16%、13.68%(P=0.585),观察组等位基因频率A、G为68.81%、31.19%,对照组为64.74%、35.26%(P=0.383);观察组IFNγ+874A/T位点AA、AT、TT型基因为48.62%、41.28%、10.10%,对照组分别为46.32%、50.53%、3.15%(P=0.102),观察组等位基因频率A、T分别为69.27%、30.73%,对照组为71.58%、28.42%(P=0.610)。观察组血浆IFNγ为(4.60±1.91)μg/L,对照组为(4.88±2.10)μg/L(P=0.318)。结论 IFNγ的基因多态性不能作为AS斑块稳定性的预警指标。Objective To study whether IFNγ＋2109A/G and ＋874A/T gene polymorphism affects the incidence of atherosclerosis and its plaque stability.Methods Two hundred and four patients admitted to our hospital from March 2014 to March 2015 were divided into unstable atherosclerotic plaque group as an observation group（n=109）and stable atherosclerotic plaque group as a control group（n=95）.Their serum IFNγgene level was measured,their IFNγ＋2109gene and IFNγ＋874gene were detected by direct sequencing of the PCR products and PCR-SSP respectively.The sequencing results of these genes were analyzed using DNAStar and GeneTool.Results The detection rate of AA,AG,GG was 46.79%,44.04%,9.17%in observation group and was43.16%,43.16%,13.68%in control group（P=0.585）.The frequency of A and G allelic genes was 68.81%,31.19% in observation group and was 64.74%,35.26% in control group（P=0.383）.The detection rate of AA,AT,TT was 48.62%,41.28%,10.10%in observation group and was 46.32%,50.53%,3.15%in control group（P=0.102）.The frequency of A and T allelic genes was 69.27%,30.73% in observation group and was 71.58%,28.42% in control group（P=0.610）.The serum IFNγgene level was 4.60±1.91μg/L in observation group and was4.88±2.10μg/L in control group（P=0.318）.Conclusion IFNγgene polymorphism cannot be used as an early warning indicator of atherosclerotic plaque stability.
