胡黄连苷Ⅱ干预脑缺血再灌注大鼠神经损伤的脑保护作用  被引量:2

摘  要:目的:探讨脑缺血再灌注大鼠神经损伤中胡黄连苷Ⅱ对脑保护的作用。方法:选取成年健康雄性大鼠40只,按照随机数字表法随机分为2组,假手术处理10只作为对照组,剩余30只为研究组,利用线栓法制作大鼠大脑动脉缺血再灌注模型,再分为3组,各10只,包括模型组、胡黄连苷Ⅱ低剂量组及胡黄连苷Ⅱ高剂量组。再灌注时及时给药,对照组与模型组同步尾注生理盐水,胡黄连苷Ⅱ低剂量组与高剂量组则分别尾注10 mg/kg、30 mg/kg胡黄连苷Ⅱ,对比几组大鼠缺血再灌注后24 h神经功能缺损评分,以及脑匀浆上清相关指标。结果:和模型组相比,胡黄连苷Ⅱ低剂量组与高剂量组在神经功能缺损评分,脑匀浆上清超氧化物歧化酶(SOD)活性、还原型谷胱甘肽水平、丙二醛(MDA)水平及一氧化氮合酶(NOS)活性等方面均有显著性差异(P<0.05)。结论:胡黄连苷Ⅱ干预脑缺血再灌注大鼠神经损伤模型有一定的脑保护作用,可能和其提高脑组织抗氧化能力与减轻氧化性损伤等有关。Objective: To explore the protective effect of picroside Ⅱ on brain of nerve injury due to cerebral ischemia reperfusion in rats. Methods: 40 healthy adult male rats were selected,and randomly divided into 2 groups. 10 rats of sham operation were taken as the control group,30 remaining rats were taken as the study group. The models of cerebral artery ischemia reperfusion in rats were made by suture-occluded method,and divided into 3 groups,10 rats in each model group,i. e. model group,low-and high-dose group of picrosideⅡ. At the time of reperfusion timely to administer: the control group and the model group were injected the normal saline synchronously in the tail,while the low-and high-dose group of picrosideⅡwere injected respectively the picrosideⅡ of 10 mg /kg and 30 mg /kg in the tail. NIHSS and the related indexes of cerebral homogenate supernatant were compared in 24 h after ischemia reperfusion in rats of several groups. Results: Compared with the model group,in the low-and high-dose group of picroside Ⅱ,there were the significant differences in NIHSS and superoxide dismutase( SOD) activity,reduced glutathione level,malondialdehyde( MDA) level and nitric oxide synthase( NOS)activity etc. of cerebral homogenate supernatant( P〈0. 05). Conclusion: Picroside Ⅱ has a certain cerebral protective effect on model of nerve injury due to cerebral ischemia reperfusion in rats,which may be related to the increase of antioxidant capacity and reduction of oxidative injury in brain tissue.
