摘 要:目的观察肿瘤易感基因101(TSG101)对肝癌细胞的增殖、侵袭、转移的影响。方法建立稳定表达TSG101的肝癌细胞株,将细胞分为转染组、空质粒组、空白对照组,通过细胞计数试剂盒(CCK-8)增殖实验观察各组细胞增殖;通过Transwel小室侵袭实验观察各组细胞侵袭能力;通过Transwel小室迁移实验观察各组细胞转移能力。结果(1)CCK-8增殖实验可见TSG101组36h后细胞增殖比例(1.93±0.45)明显高于空白对照组(1.28±0.51)与空质粒组(1.31±0.29),且差异有统计学意义(P〈0.05);空白对照组与空质粒组比较差异无统计学意义(P〉0.05);(2)Transwell小室细胞侵袭实验:TSG101组[(125.0±10.3)个]细胞穿透个数明显高于空白对照组[(83.0±8.7)个]与空质粒组[(85.0±9.2)个],且差异有统计学意义(P〈0.05);空白对照组与空质粒组比较差异无统计学意义(P〉0.05);(3)Transwell小室细胞迁移实验:TSG101组[(88.0±7.0)个]穿透细胞个数高于空白对照组[(59.0±6.8)个]与空质粒组[(53.0±7.7)个],且差异有统计学意义(P〈0.05);空白对照组与空质粒组比较差异无统计学意义(P〉0.05)。结论高表达TSG101蛋白的肝癌细胞增殖、侵袭、转移能力均增强。Objective To study of the effects of tumor susceptibility gene 101 (TSG101) on pro- liferation, invasion and metastasis of liver cancer cells. Methods To establish stable expression of TSG101 liver cancer cell lines, then cells can be divided into transfection group, the empty plasmid group, blank control group, through the cell counting kit - 8 ( CCK - 8 ) multiplication experiment observation group cell proliferation; and by Transwell chambers attacking invasion ability and migration experiment a- bility to observe each cell metastasis. Results CCK - 8 multiplication experiment found TSGI0I group and 36 h after cell proliferation rate (1.93 ± 0. 45 ) significantly higher than the blank control group ( 1.28 ± 0. 51 ) and the empty plasmid group ( 1.31 ± 0. 29), and the difference was statistically significant (P 〈0. 05). Blank control group compared with the empty plasmid group had no statistical significance (P〉0. 05); (2) the transwell Chambers cell invasion experiment: TSG101 group cells through number (125. 0 ± 10. 3) significantly higher than the blank control group (83. 0 ± 8. 7) and the empty plasmid group (85.0 ±9. 2), and the difference was statistically significant (P 〈0. 05). Blank control group com- pared with the empty plasmid group had no statistical significance; (3) transwell Chambers cell migration experiment: TSG101 group penetrate the cell number (88. 0 ± 7. 0) is higher than the blank control group (59. 0 ± 6. 8 ) and the empty plasmid group (53. 0 ± 7.7 ), and the difference was statistically significant ( P 〈 0. 05 ). Blank control group compared with empty plasmid groups has no statistical significance. Con- clusion High expression of TSG101 protein of liver cancer cell proliferation, invasion and metastasis abili- ty are enhanced.
