摘  要:目的探讨地塞米松(DXM)与甲基强的松龙(MP)对C6胶质瘤大鼠放射性脑损伤后脑水肿程度的影响以及与表皮生长因子受体1(erb-B1)基因转录水平的关系。方法雄性Wistar大鼠100只,体质量250~300g,随机分成5组,每组20只,分别为接受照射并接受激素治疗的30mr/kg甲基强的松龙组(A)、10mg/kg甲基强的松龙组(B)、5mg/kg地塞米松组(c),接受照射但未接受激素治疗的实验对照组(D),未接受照射和激素治疗的正常对照组(E)。各组均颅内种植肿瘤,A、B、C、D组于种植15d后均给予^60Co照射,同时自照射前3d开始静脉注射激素或生理盐水,直至照射后6d,种植肿瘤后21d取大鼠脑组织测含水量和erb-B1转录水平。结果脑组织含水率分别为A组:0.7588±0.0159;B组:0.7669±0.0174;C组:0.7965±0.0197;D组:0.8283±0.0216;E组:0.7270±0.0113。D、E组分别与A、B、C组比较,差异有统计学意义(P〈0.05);D、E两组间比较,差异有统计学意义(P〈0.01);C组分别与A、B组比较,差异有统计学意义(P〈0.05);A、B两组间比较,差异无统计学意义(P〉0.05)。erb-B1转录水平:A组:0.7391±0.0415;B组:0.7410±0.0344;C组:0.7754±0.0546;D组:0.8021±0.0558;E组:0.7156±0.0592。D、E组分别与其他各组比较,差异有统计学意义(P〈0.05);D、E两组间比较,差异有统计学意义(P〈0.01);C组分别与A及B组比较,差异有统计学意义(P〈0.05);A组与B组比较,差异无统计学意义(P〉0.05)。结论甲基强的松龙与地塞米松均可通过抑制erb-B1基因的转录的方式减轻放射性脑水肿(RCE),而且甲基强的松龙效果更好。Objective To study the effect of methylprednisolone (MP) and dexamethasone (DXM) on radioactive cerebral edema and the transcription level of epidermal growth factor receptor 1 (erb- B1 ). Methods One hundred male Wistar rats were divided into 5 groups randomly (n = 20). Group A was irradiated in combination with 30 mg/kg MP, group B was irradiated in combination with 10 mg/kg MP, group C was irradiated in combination with 5 mg/kg DXM, group D was irradiated without glucoeorticoid ( GC), and group E served as normal control. Glioma cells were planted into brains of rats in groups A, B, C, D and E, and the animals in groups A, B, C, and D were given irradiateion of 60Co after 15 days. All groups were give GC or normal saline 3 days before and 6 days after irradiation. The wa- ter content of the brains and the transcription level of erb - B1 were measured 6 days after irradiation. Re- sets The water eontem of the rats brain : A:0.7588±0.0159, B:0.7669±0.0174, C:0.7965± 0. 019 7, D: 0. 828 3 ±0. 021 6, E: 0. 727 0 ±0. 011 3. There was significant difference between group D, E and group A, B, C ( P 〈 0. 05 ) , group D and group E ( P 〈 0. 01 ), group C and group A, B ( P 〈 0. 05) ; There were no significant difference between group A and B (P 〉 0. 05 ). The level of erb - B1 mRNA: A: 0.739 1 ±0.041 5, B: 0.741 0±0.0344, C: 0.7754±0.0546, D: 0.802 1 ±0.055 8, E: 0.715 6±0.0592. There were significant difference between group D, E and group A, B, C (P〈 0. 05), group D and group E (P 〈0. 01 ), group C and A, B (P 〈0. 05). There were no significant differ- ence between group A and B (P 〉 0. 05 ). Conclusion MP and DXM can both protect the radiation - in- duced injury and brain edema changes by inhibiting the transcription level of erb - B1 mRNA in rats, and the MP is more efficient than DXM.
