摘 要:目的观察半乳糖凝集素-1(Galectin-1)在结肠腺癌中的表达及其临床意义。方法采用原位杂交技术和免疫组织化学方法检测75例结肠腺癌组织、43例癌旁不典型增生组织及75例正常结肠黏膜上皮组织中Galeetin-1mRNA及蛋白的表达,并分析其与临床病理学参数之间的关系。结果原位杂交的结果表明,结肠腺癌组织中Galeetin-1mRNA表达的阳性率(84.00%)明显高于癌旁不典型增生组织(34.88%)和正常结肠上皮组织(22.67%),三者之间差异有统计学意义(P〈0.05)。免疫组织化学结果表明,结肠腺癌组织中Galeetin-1蛋白表达的阳性率(80.00%)明显高于癌旁不典型增生组织(30.23%)和正常结肠上皮组织(21.33%),三者之间差异有统计学意义(P〈0.05)。Galectin-1mRNA和Galeetin-1蛋白的表达与组织学分级和结肠腺癌的浸润深度无关(P〉0.05),但是与淋巴结转移和TNM分期明显相关(P〈0.05)。Galectin-1mRNA和蛋白表达的相关分析结果表明,Galectin-1mRNA及蛋白在结肠腺癌组织中的表达强度呈现出正相关(r=0.600,P〈0.01)。结论Galectin-1mRNA和蛋白在结肠腺癌中的表达显著高于癌旁不典型增生组织和正常结肠黏膜组织,其高表达可能在结肠腺癌发生发展中发挥重要作用。Galectin-1可能是一种判定结肠腺癌恶性程度极其重要的分子标记。Galectin-1mRNA和蛋白在结肠腺癌组织中的表达呈正相关。Objective To observe the expression and clinical significance of Galectin - 1 in hu- man colon adenocarcinoma. Methods Expression of Galectin- 1 mRNA and protein was detected by in situ hybrid method and immunohistochemistry method in 75 cases of colon adenocarcinoma tissues, 43 ca- ses of mucosa adjacent to cancer, and 75 cases of normal colon mucosa, and correlations between expres- sion of Galectin - 1 and clinicopathological features was analyzed. Results The results of in situ hybrid revealed that the expression of Galectin - 1 mRNA in colon adenocarcinoma tissues ( 84. 00% ) was signifi- candy higher than those of mucosa adjacent to cancer (34. 88% ) and normal esophageal mucosa epithelial tissue (22. 67% ), there were significant differences in Galectin - 1 mRNA expression among three groups (P 〈 0. 05). The results of immunohistochemistry showed that Galectin - 1 protein in colon adenocarcino- ma tissues (80. 00% ) was significantly higher than those of mucosa adjacent to cancer (30. 23% ) and nor- mal colon mucosa epithelial tissue (21.33%), there were significant differences in Galectin - 1 protein ex- pression among three groups (P 〈0. 05). Expression of Galectin - 1 mRNA and Galectin - 1 protein in colon adenocarcinoma tissue is not related histologic grade and invasion depth ( P 〉 0. 05 ), but closely related to TNM staging and lymph node metastasis (P 〈 0. 05 ). The results of correlation analysis of Galectin - 1 mRNA and protein expressions demonstrated that the expression of Galectin - 1 mRNA and protein in colon adenocarcinoma tissues displayed positive correlation ( r = 0. 600, P 〈 0. 01 ). Conclusion Expressions of Galectin - 1 mRNA and protein in colon adenocareinoma tissues are significantly higher than those in muco- sa adjacent to cancer and normal colon epithelium tissues, and Galectin - 1 at high level plays an important role in occurrence and development of colon adenocarcinoma.
