B细胞淋巴瘤基因-2、Twist家族碱性螺旋-环-螺旋转录因子1转录因子在舌鳞癌中调控上皮-间充质转化的研究

摘　　要：目的探讨B细胞淋巴瘤基因-2（bcl-2）、Twist家族碱性螺旋-环-螺旋转录因子1（Twistl）在舌鳞癌中的表达及对上皮-间充质转化（EMT）调控。方法应用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶（SP）法检测bcl-2、Twistl及EMT相关因子在82例舌鳞癌及24例癌旁组织中的表达，分析bcl-2、Twistl及EMT相关因子与舌鳞癌临床病理特征的关系。Westernblot法检测舌鳞癌细胞株Tb3．1中bcl-2、Twistl及EMT相关因子表达水平；免疫共沉淀证明bcl-2、Twistl的相互作用。结果bcl-2、Twistl在舌鳞癌组织中的表达明显高于癌旁组织，82例癌组织中48例（58．64％）bcl-2、46例（56．10％）Twistl表达呈阳性表达，明显高于癌旁组织[bcl-2：6例（25．00％），P〈0．05，Twistl：4例（16．67％），P〈0．05]；bcl-2、Twistl、神经钙黏素、波形蛋白在中低分化组及淋巴转移组中阳性表达高于在高分化组及非淋巴结转移组中的表达，在中低分化组中的表达分别为28例（70．00％）、28例（70．00％）、30例（75．00％）、30例（75．00％），在淋巴结转移组中的表达分别为29例（72．50％）、29例（72．50％）、29例（72．50％）、31例（77．50％），钙黏着蛋白则与之相反，分别为15例（37．50％）、16例（40．00％）。且Twistl与bcl．2、神经钙黏素、波形蛋白的表达呈正相关（相关系数分别为0．303、0．249、0．348，P〈0．05），与钙黏着蛋白的表达呈负相关（相关系数为-0．547，P〈0．05）；免疫共沉淀结果显示bcl-2、Twistl间可互相作用。结论bcl-2和Twistl在舌鳞癌中高表达，两者形成复合体可能参与调控舌鳞癌EMT过程。Objective To sutdy the expression of B cell lymphoma/leukemia -2 （bcl -2）, twist family basic helix - loop - helix transcription factor 1 （ Twistl ） in oral squamous cell carcinoma and its modulating epithelial -mesenchymal transition （EMT）. Methods Immunobistochemistry was used to de- tect the bcl -2, Twistl and EMT related proteins expression among 82 OSCC samples and 24 paracancer- ous samples. The relationship between the bcl - 2, Twistl and the EMT biomarkers was analyzed. Western blotting were used to observe the bcl - 2, Twistl and EMT related proteins expression. Immunoprecipitation were used to assessed interaction between bcl -2 and Twistl. Results bcl -2, Twistl expression in oral squamous cell carcinoma was significantly higher than the adjacent tissues. Of 82 cases OSCC, bcl - 2 overexpression were found in 48 cases （58.64%） and Twistl overexpraeeion were in 46 cases （ 56. 10% ）, which were significantly higher than the adjacent tissues [ bcl - 2 ： 6 cases （ 25.00% ）, P 〈0. 05 ,Twist1：4 cases （16. 67% ）,P 〈 0. 051. In poorly differentiated and lymph node metastasis group, the expression of bcl - 2, Twist!, N - cadherin, Vimentin were higher than those in high differentiation and non - lymph node metastasis group. Twistl showed a positive correlation with bcl - 2, N - cadherin, Vimentin （ correlation coefficients were 0. 303,0. 249, 0. 348 respectively, P 〈 0.05 ）, but negatively cor- related with E - cadherin （ correlation coefficient of - 0. 547, P 〈 0. 05 ）. Immunoprecipitation showed that bcl -2, Twistl can interact with each other. Conclusion bcl -2 and Twistl overexpressed in oral squa- mous cell carcinoma and form a complex, bcl -2/Twistl complex might play a critical role in modulating EMT.
