摘  要:目的:探讨SATB1与前列腺癌增殖及侵袭发生的关系。方法:体外培养前列腺癌细胞系(LNCa P、PC3、DU145),MMT法检测其增殖能力;Tanswell细胞侵袭实验检测其侵袭能力;RT-PCR检测其SATB1 mRNA的表达情况;Western blot检测其SATB1蛋白质的表达。结果:前列腺癌细胞PC3和DU145的增值能力及侵袭能力比LNCaP强(P<0.01),PC3和DU145细胞中SATB1 mRNA及蛋白质表达水平均显著高于LNCaP细胞(P<0.01)。结论:SATB1在前列腺癌表达水平可能与前列腺癌的增殖和侵袭正相关。Objectives: To observe the expression of SATB1 in different prostate cancer cell lines,and explore its possible relation with prostate cancer proliferation and invasion potential. Methods: The proliferation ability of LNCa P,PC3 and DU145 cell were detected by MTT. The transwell chamber was used to confirm the invasive potency of LNCa P,PC3 and DU145 cell lines. The expressions of the SATB1 protein and mRNA in these prostate cancer cells with different metastatic potentials were detected by Western blotting and RT- PCR,respectively.Results: Compared with the LNCa P cell line,the proliferation ability of PC3 and DU145 cell lines was higher( P〈0. 01). The SATB1 protein and mRNA highly expressed in the PC3 and DU145 cell line that was well- known with a high metastatic potential,but not in the LNCa P cells with low metastatic potentials( P〈0. 01). Conclusion:SATB1 deficient expression may play an important role in the proliferation and invasion of advanced prostate cancer.
