摘  要:目的构建一种基于分子信标与核酸适体的荧光传感器,用于三磷酸腺苷分子的选择性检测。方法将三磷酸腺苷的核酸适体序列与分子信标结合形成新的荧光探针,利用腺苷和分子信标互补DNA的竞争,引发分子信标的构型转换,从而产生可检测的荧光响应信号实现检测腺苷的目的。结果荧光响应信号随着腺苷浓度的增加而减弱。在3.70×10^(-6)~5.56×10^(-4)mol/L的浓度范围内,腺苷浓度的对数和响应信号成线性关系,相关系数为0.9974。加标回收率为92.0%~110.0%,相对标准偏差为0.68%~1.85%(n=3)。结论该方法耗时较少、步骤简单,可满足当前生化分析快速而精确的分析要求。Objective To construct a fluorescence sensor based on molecular beacon and aptamer of nucleic acid for the selective detection of adenosine triphosphate.Methods The novel fluorescent probe was designed by combining aptamer of adenosine triphosphate with molecular beacon.Configuration transformation of the probe could occur when adenosine and complementary DNA competitively hybridized with it.And then a detectable fluorescent response signal was generated so as to realize the purposes of detection for adenosine.Results The fluorescent intensity decreased with the increase of adenosine concentration.The fluorescent intensity was linearly dependent on the adenosine concentration in the range of 3.70×10^(-6)~5.56×10^(-4) mol/L with the correlation coefficient of 0.997.The recoveries were 92.0%~110.0%,and the relative standard deviations(RSD) were 0.68%~1.85%(n=5).Conclusion The proposed method is time-saving and simple,which can meet the rapid and precise requirements of the current biochemical analysis.
