摘 要:目的:探讨不同浓度NaHS(硫化氢供体)对人卵巢癌SKOV3细胞增殖的影响。方法:体外培养人卵巢癌SKOV3细胞,分为正常对照组、NaHS(25μmol/L)组、NaHS(50μmol/L)组、NaHS(75μmol/L)组、NaHS(100μmol/L)组,采用MTT法检测各组干预48h后SKOV3细胞增殖情况,流式细胞术检测各组干预48h后SKOV3细胞周期分布情况。结果:与正常对照组比较,NaHS(25μmol/L)组、NaHS(50μmol/L)组、NaHS(75μmol/L)组、NaHS(100μmol/L)组SKOV3细胞抑制率均显著增加,G0/G1期细胞均显著增加,S期细胞均显著减少,细胞增殖指数均明显降低;与NaHS(25μmol/L)组比较,NaHS(50μmol/L)组、NaHS(75μmol/L)组、NaHS(100μmol/L)组SKOV3细胞抑制率均显著增加,G0/G1期细胞均显著增加,S期细胞均显著减少,细胞增殖指数均明显降低;与NaHS(50μmol/L)组比较,NaHS(75μmol/L)组、NaHS(100μmol/L)组SKOV3细胞抑制率均明显降低,G0/G1期细胞均明显减少,S期细胞均明显增加,细胞增殖指数均明显降低;而NaHS(75μmol/L)组在细胞抑制率、G0/G1期细胞比例、S期细胞比例、细胞增殖指数等方面与NaHS(100μmol/L)组无明显差异。结论:硫化氢可抑制人卵巢癌SKOV3细胞的增殖,并呈明显浓度依赖性。Objective:To investigate the effects of hydrogen sulfide on the proliferation of the human ovarian carcinoma cells SKOV3.Methods:The cultured human ovarian carcinoma cells SKOV3 were divided into the normal control group,hydrogen sulfide(25μmol/L)group,hydrogen sulfide(50μmol/L)group,hydrogen sulfide(75μmol/L)group and the hydrogen sulfide(100μmol/L)group,the cultured human ovarian carcinoma cells were treated for 48 hours.The cells proliferation was measured by MTT assay.The cells cycle was measured by flow cytometry.Results:Compared with the normal control group,the Cell inhibition rate in the hydrogen sulfide(25μmol/L)group,hydrogen sulfide(50μmol/L)group,hydrogen sulfide(75μmol/L)group and the hydrogen sulfide(100μmol/L)group were increased.All the cells of G1 phase were significantly increased and the cells of S phase were significantly decreased.Compared with the hydrogen sulfide(25μmol/L)group,the Cell inhibition rate in the hydrogen sulfide(50μmol/L)group,hydrogen sulfide(75μmol/L)group and the hydrogen sulfide(100μmol/L)group were significantly increased.All the cells of G1 phase were significantly increased and the cells of Sphase were significantly decreased.Compared with the hydrogen sulfide(50μmol/L)group,the cell inhibition rate in the hydrogen sulfide(75μmol/L)group and hydrogen sulfide(100μmol/L)group were significantly increased.All the cells of G1 phase were significantly increased and the cells of S phase were significantly decreased.Conclusion:The hydrogen sulfide can inhibit the human ovarian carcinoma cells proliferation in aconcentration-dependent manner.
