摘  要:目的探讨非小细胞肺癌(NSCLC)淋巴结中细胞角蛋白19(CK19)、肺组织特异性X蛋白(LunX)、上皮癌相关基因(KS1/4)的表达与NSCLC微转移的关系。方法选择经过9个月随访的52例NSCLC患者,分为转移组和无转移组。提取淋巴结标本的总RNA,反转录为cDNA,采用荧光定量实时PCR检测其中CK19、LunX、KS1/4的mRNA表达水平。结果总RNA通过紫外线分光光度仪检测RNA纯度,OD260/280值为1.650~2.1,RNA浓度为560~1420mg/L。经过随访,52例研究病例中发生转移或复发共18例,转移复发率为34.6%。其中转移组淋巴结中CK19、LunX、KS1/4的mRNA表达均高于无转移组(P值均〈0.05)。结论CK19、LunX、KS1/4可以作为预测NSCLC转移或复发的指标,辅助诊断NSCLC微转移。Objective To explore the relationship between micrometastasis and CK19, LunX and KS1/4 in non-small cell lung cancer (NSCLC) patients. Methods After nine monthsI following-up, all the 52 patients were divided into metastasis group and non-metastasis group. 20 patients with pulmonary benign diseases were chosen to be control group. Total RNA was extracted from lymph nodes, mRNA was transcribed reversely into cDNA. The expressions of genes including CK19, LunX and KS1/4 were detected according to fluoregenic quantitative real time polymerase chain reaction. Results The concentration of total RNA after extraction was 560-1 420 mg/L, and OD260/280 was 1. 650-2.1. The rate of metastasis or recurrent was 34.6% after following-up. The expressions of CK19 mRNA, LunX mRNA,KS1/4 mRNA in lymph nodes of metastasis group were higher than those of non-metastasis group (all P 〈0.05).Conclusions CK19, LunX and KS1/4 can be used as sensitive markers to predict metastasis or recurrent of NSCLC, make auxiliary diagnosis for micrometastasis.
