摘 要:目的观察骨碎补总黄酮对牙周炎大鼠龈沟液骨钙素(OC)和牙槽骨骨密度的影响,探讨其治疗牙周炎的机制。方法成功建立SD大鼠牙周炎模型之后,使用完全随机分组方法将大鼠分为正常组、牙周炎组、骨碎补总黄酮组,每组18只。建模4周后分别灌服生理盐水和骨碎补总黄酮240mg/(kg·d)。各组大鼠分别在建模后第4,6,8周各处死6只。放射免疫分析法测定龈沟液OC水平,显微CT测定大鼠牙槽骨骨密度。结果牙周炎组龈沟液OC水平显著高于正常组和骨碎补总黄酮组(P<0.05);骨碎补总黄酮组龈沟液OC水平在6周和8周时显著低于牙周炎组(P<0.05),8周时与正常组龈沟液OC水平相比差异无统计学意义(P>0.05)。牙周炎组牙槽骨骨密度显著低于正常组和骨碎补总黄酮组(P<0.05);骨碎补总黄酮组牙槽骨骨密度在6和8周时显著高于牙周炎组(P<0.05)。结论骨碎补总黄酮可降低牙周炎大鼠龈沟液骨钙素水平,增加牙槽骨骨密度,对大鼠牙周炎具有一定治疗作用。Objective To observe the effect of Rhizoma Drynariae Flavone on the sulcular fluid level of osteocalcin(OC)and alveolar bone mineral density during experimental periodontitis for its therapy mechanism.Methods The model of experimental periodontitis was established by means of silk ligatures around and high-carbohydrate diet.Then the animals were randomly assigned to three groups and received daily oral treatment.Four weeks after the ligure,Rhizoma Drynariae Flavone was given to rats of treatment group at a dose of 240mg/kg/d by gastric injuction,and the same amount of saline was given to rats of normal control and periodontitis group for same period.Six animals in each group were randomly selected and sacrigiced at 4,6and 8weeks.OC was detected by radio immunoassay,alveolar bone density was detected by micro-computed tomographic.Results The level of OC in experimental periodontitis group was significantly higher than that in control group and treatment group(P〈0.05),and the level of OC in treatment group was significantly lower than that in periodontitis group at week 6and 8(P〈0.05).There were no statistical significantly differences between control group and treatment group at week 8(P〈0.05).The alveolar bone mineral density in periodontitis group was significantly lower than that in control and treatment group(P〈0.05).The level of alveolar bone mineral density in treatment group was significantly higher than that in periodontitis group at weeks 6and 8(P〈0.05).Conclusion Rhizoma Drynariae Flavone reduces the level of osteocalcin in the sulcular fluid,increases aoveolar bone mineral density.Rhizoma Drynariae Flavone could play an important role in treating periodontitis.
