摘　　要：目的探讨提取石蜡包埋胃癌组织中DNA的最佳方法。方法用蛋白酶K消化氯化钠盐析方法、蛋白酶K消化酚氯仿抽提方法、试剂盒方法及改良试剂盒方法,提取石蜡包埋胃癌组织中DNA,进行PCR扩增,应用凝胶电泳成像检出率(检出率=检出数/总例数)比较4种方法提取DNA质量的情况。结果改良试剂盒方法,凝胶电泳成像检出率95.5%(151/158),分别与蛋白酶K消化氯化钠盐析方法15.3%(4/26)、蛋白酶K消化酚氯仿抽提方法8.3%(1/12)、试剂盒方法19.2%(11/57)凝胶电泳成像检出率比较差异有统计学意义(P<0.05)。结论 4种提取DNA方法中改良试剂盒方法,是提取石蜡包埋胃癌组织中DNA的最佳方法。Objective： The investigation on extracting the high quality DNA from c cancer tissues Methods： Using the following methods, for examples, proteinase K digestion and sodium chloride salting-out, proteinase K digestion and phenol-chloroform extraction, the extractionkitand its improved kit. DNA was extracted from paraffin- embeddedgastric cancer tissues, and PCR amplification and gel electrophoresis were performed to confirm the quality DNA. Results： DNA that was extracted tric cancer tissues following those methods, the extraction improved kit, proteinase K digestion and sodium chloride salting-out method, proteinase K digestion and phenol chloroform extraction, and the extraction kit. Respectively, the detection rate was 19.2% （11/57） , 15.3% （4/26） , 8.3% （1/12） , and 19.2% （11/57） . Importantly, the difference was statistically significant （P〈0.05） , the extraction improved kit compared with other three methods. Conclusion： The results confirmed that utilizing improved DNA Extraction Kit can obtain the high quality template DNA from paraffin-embeddedgastric cancer tissues.
