摘 要:目的探讨多种细胞因子诱导的杀伤细胞(CIK)回输对Ⅳ期结直肠癌患者外周血活化T细胞和中央型记忆T细胞的影响,初步分析CIK细胞的抗肿瘤机制。方法选择我院2014年10月—2015年10月收治的Ⅳ期结直肠癌患者47例,均以静脉输注的方式予3个周期CIK细胞治疗,治疗前和治疗结束后检测外周血中活化T细胞(CD38+HAL-DR+)及中央型记忆T细胞(CD45RO+CD62L+)的占比并进行比较。结果所有患者治疗前均行CIK细胞的制备,培养成熟的CIK细胞数量为(4.87±2.02)×108,其中CD3+CD8+T细胞和CD3+CD56+NK细胞占比分别为(74.31±15.45)%和(32.33±13.57)%。治疗前患者外周血中活化T细胞及中央型记忆T细胞占比分别为(15.35±8.46)%和(20.13±10.53)%,治疗后分别增高至(38.73±14.72)%和(41.18±10.67)%,差异有统计学意义(P<0.05)。结论 CIK细胞可通过扩增中央型记忆T细胞及激活T细胞来重建患者的抗肿瘤免疫,进而发挥抗肿瘤效应。Objective To explore the impact of CIK treatment on percentage of activated T cells and central memory T cells in patients with stage Ⅳ colorectal cancer,and analyze the mechanisms of anti-tumor effect of CIK cells. Methods47 patients with stage Ⅳ colorectal cancer undergoing CIK treatment in our hospital during October 2014 and October 2015 were enrolled in this study. Percentage of activated T cells( CD38 + HLA-DR +) and central memory T cells( CD45 RO +CD62L +) in the peripheral blood before and after CIK treatment were evaluated and compared respectively. Results( 4. 87 ± 2. 02) × 108 CIK cells were obtained through preparation,and the percentage of CD3 + CD8 + T cells and CD3 +CD56 + NK cells in CIK cells were( 74. 31 ± 15. 45) % and( 32. 33 ± 13. 57) % respectively. Percentage of activated T cells and central memory T cells in the peripheral blood before CIK treatment were( 15. 35 ± 8. 46) % and( 20. 13 ± 10. 53) % respectively. After CIK treatment,Percentage of activated T cells and central memory T cells were( 38. 73 ± 14. 72) % and( 41. 18 ± 10. 67) % respectively. There was significant difference in percentage of activated T cells and central memory T cells in the peripheral blood before and after CIK treatment( P〈0. 05). Conclusion CIK can restore the anti-tumor immunity through inducing memory T cells and activating T cells with anti-tumor efficacy.
