摘 要:目的观察1,25-二羟基维生素D3[1,25(OH)2Vit D3]对高氧环境中人肺腺癌A549细胞凋亡的影响,并探讨其机制。方法取对数生长期A549细胞,随机分为5组。A组于37℃、5%CO2饱和湿度培养箱中培养,不处理;B组于高氧环境中培养,不处理;C、D、E组于高氧环境中培养,并分别加入终浓度为0.1、1、10μmol/L的1,25(OH)2Vit D3处理48 h。流式细胞仪检测凋亡细胞数并计算细胞凋亡指数,Western blot法检测细胞中的Bcl-2、Bax蛋白。结果 A、B、C、D、E组细胞凋亡指数分别为2.45±0.91、40.83±4.96、42.65±4.85、63.43±5.61、76.47±5.94,A组与其他四组比较P均<0.05,B、C组比较P>0.05,D、E组与B、C组比较P均<0.05,D、E组比较P<0.05。与A组比较,B、C、D、E组Bcl-2蛋白表达及Bcl-2/Bax值均下降,而Bax蛋白表达均增高,P均<0.05。B、C组Bcl-2、Bax蛋白表达及Bcl-2/Bax值比较,P均>0.05。与B、C组比较,D、E组Bcl-2蛋白表达及Bcl-2/Bax值下降,而Bax蛋白表达增高,P均<0.05。与D组比较,E组Bcl-2蛋白表达及Bcl-2/Bax值下降(P均<0.05),而Bax蛋白表达变化不明显(P>0.05)。结论 1,25(OH)2Vit D3可促进高氧环境中肺腺癌A549细胞凋亡,可能与其调节Bcl-2/Bax平衡有关。Objective To observe the effects of 1,25-dihydroxyvitamin D3[1,25( OH)2Vit D3]on the apoptosis of human lung adenocarcinoma A549 cells in the high oxygen environment and to explore its mechanism. Methods A549 cells in the logarithmic phase were randomly divided into five groups: groups A,B,C,D and F. Cells in the group A were cultured at 37 ℃,5% CO2 humidified incubator,and were not processed; cells in the group B were cultured in high-oxygen environment,and were not processed; cells in the groups C,D and E were cultured in high oxygen environment,and then were treated with 0. 1,1 and 10 μmol / L of 1,25( OH)2Vit D3 for 48 h. Apoptotic cells were detected by flow cytometry,and the Bcl-2,Bax protein was detected by Western blotting. Results The apoptosis indexes of groups A,B,C,D and E group were 2. 45 ± 0. 91,40. 83 ± 4. 96,42. 65 ± 4. 85,63. 43 ± 5. 61 and 76. 47 ± 5. 94,respectively. Significant difference was found between group A and groups B,C,D and E,all P 0. 05; no significant difference was found between group B and group C,P 0. 05; significant difference was found between groups D,E and groups B,C,all P 0. 05; and significant difference was found between group D and group,P 0. 05. Compared with group A,the Bcl-2 protein expression and Bcl-2 / Bax value were decreased,but the Bax protein expression was increased in the groups B,C,D and E( all P 0. 05). No significant difference was found in the Bcl-2,Bax protein expression and Bcl-2 / Bax value between group B and group C,all P 0. 05. Compared with group B and group C,the Bcl-2 protein expression and Bcl-2 /Bax value were decreased,but the Bax protein expression was increased in the groups D and E( all P 0. 05). Compared with group D,the Bcl-2 protein expression and Bcl-2 / Bax value were decreased( all P 0. 05),and the Bax protein expression did not change in the group E( P 0. 05). Conclusion 1,25( OH)2Vit D3 may promote the apoptosis of lung adenocarcinoma A549 cells in a high oxygen environment by regulating Bcl-2
