摘  要:目的观察miR-124对非小细胞肺癌(NSCLC)细胞迁移和侵袭的调控作用,并探讨其作用机制。方法取NSCLC细胞系A549,分为对照组(转染microRNA scramble)、miR-124组(转染miR-124 mimics)和miR-124+Smad4组(转染miR-124 mimics和pc DNA3.1-Smad4),采用细胞划痕实验和Transwell小室基质渗透实验观察三组细胞迁移和侵袭能力,蛋白印迹法测定E-Cadherin、N-Cadherin、Vimentin蛋白表达。将A549分为三组,野生型质粒组、突变型质粒组及阴性对照组,采用荧光素酶法鉴定miR-124与Smad4的靶向关系。结果 miR-124组的划痕愈合率低于miR-124+Smad4组和对照组,侵袭细胞数少于其他两组(P均<0.01)。miR-124组E-cadherin、Vimentin蛋白相对表达量高于miR-124+Smad4组和对照组,N-cadherin蛋白相对表达量低于其他两组(P均<0.05)。野生型质粒组萤光素酶活性显著低于阴性对照组和突变型质粒组,证实miR-124与Smad4之间存在靶向关系。结论miR-124通过抑制Smad4基因表达抑制上皮间质转化,进而抑制NSCLC细胞迁移和侵袭。Objective To observe the regulating effect of miR-124 on the cell migration and invasion of non-small-cell lung cancer( NSCLC),and to investigate its mechanism. Methods A549 cells were divided into three groups. The control group was transfected with microRNA scramble,the miR-124 group with miR-124 mimics and the miR-124 + Smad4 group with miR-124 mimics and pc DNA3. 1-Smad4. The cell migration and invasion was measured by scratch test and Transwell invasion testing system,respectively. The protein expression levels of E-cadherin,N-cadherin and Vimentin were measured by Western blotting. The A549 cells were divided into three groups: wild type plasmid group,mutant type plasmid group and negative control group. The relationship between miR-124 and Smad4 was identified by luciferase activity.Results The wound healing rate of the miR-124 group was significantly lower than that of the control group and miR-124 +Smad4 group; the invasive cells were also less than those of the other two groups( all P 0. 01). The protein levels of Ecadherin and Vimentin in the miR-124 group were higher than those of the miR-124 + Smad4 group and the control group;but the expression level of N-cadherin in the miR-124 group was lower than that of the other two groups( all P 0. 05).The luciferase activity in the wild type plasmid group was significantly lower than that of the mutant type plasmid group and negative control group,which indicated there was some target relationship between miR-124 and Smad4. Conclusion mir-124 inhibits the migration and invasion of lung cancer cells by suppressing Smad4 gene expression and epithelial-mesenchymal transition.
