摘 要:目的检测小鼠活化巨噬细胞真菌性角膜炎中的人树突状细胞源性C型凝集素样受体-1(Dectin-1)受体、白细胞介素-17(IL-17)及白细胞介素-12(IL-12)的表达水平。方法分别向小鼠角膜创口内注入乳胶微粒液或PBS液构成实验组与对照组。于感染后第1,3,5,7,9d,裂隙灯显微镜下观测各组小鼠真菌性角膜炎特点,H-E染色观测其病理特征;通过Realtime-PCR检测小鼠角膜中Dectin-1受体、IL-17及IL-12基因mRNA的表达。结果 (1)实验组角膜浸润混浊,于第7天多发穿孔,未发现新生血管;对照组角膜混浊,于第5天广泛分布新生血管,未发现穿孔。(2)实验组角膜大量炎症细胞汇集,第5天基质细胞排列紊乱,破坏严重。(3)Realtime-PCR结果表明感染后第3,5,7,9d,实验组Dectin-1受体、IL-17、IL-12基因mRNA的表达量显著高于对照组(P<0.05)。结论 Dectin-1受体、Th1及Th17细胞免疫可能在角膜抵御真菌感染的机制中发挥重要作用。Objective To investigate expression level of c-type lectin-1receptor(dectin-1),interleukin-17(IL-17)and interleukin-12(IL-12)in mice afflicted with fungal keratitis after the activation of macrophages. Methods Prior to the infection of Fusarium solani,BALB/c mice received subconjunctival injection of latex beads or liposomes containing phosphate-buffered saline(PBS-LIP)in the corneas to establish the experimental group or the control group,respectively. After infection,the morphological features of fungal keratitis were examined under the slit-lamp and microscope at day 1,3,5,7,9and the histopathological changes were analyzed by H-E staining. The cytokine mRNA levels of IL-12,IL-17,and dectin-1in the cornea were measured by real-time PCR. Results The experimental group presented heavy inflammation,diffuse opacity,zero neovascularization,and corneal perforation at day 7;the control group showed diffuse opacity,extensive neovascularization at day 5,but no corneal perforation. Histopathologic examination of corneas found in the experimental group marked dense inflammatory cell infiltration and damage of the corneal stroma at day 5;The mRNA expressions of dectin-1,IL-17,and IL-12 in the experimental group were markedly enhanced at day 3,5,7,9when compared with those in the control group. Conclusion Dectin-1,IL-17,and IL-12 may play an important role in host defense against corneal infection of F.solani.
