摘 要:目的探讨miR155在胃腺癌发病中的作用及可能机制。方法收集2013年1月-2014年12月在南京医科大学附属苏州医院确诊的胃腺癌患者30例,收集临床资料,抽取外周血5 ml分离血浆,检测外周血中miR155和胰岛素样生长因子1(insulinlike growth factor,IGF-1),同时留取胃腺癌手术标本及癌旁正常组织,检测组织内miR155和IGF-1,分析它们的相关性。20名健康体检者外周血作为对照。结果胃腺癌患者外周血及组织中miR-155的表达明显增高,并随着临床分期的增高而进一步增高。胃腺癌患者外周血及组织中IGF-1明显增高,表达水平与肿瘤侵袭程度有关。miR155与IGF-1具有相关性。结论 miR155参与胃腺癌的发病,可能通过调控IGF-1/IGF-1R信号通路来促进胃腺癌生长。Objective To explore the role and mechanism of miR155 in the pathogenesis of gastric adenocarcinoma. Methods The clinical data of 30 gastric adenocarcinoma patients in Suzhou Hospital Affiliated to Nanjing Medical Uni- versity from Jan. 2013 to Dec. 2014 were collected. 5 ml peripheral blood were extracted to detect miR155 and insulin- like growth factor-1 (IGF-1). Meanwhile, gastric adenocarcinoma tissue and adjacent non-tumor tissue specimens were collected to detect miR155 and IGF-1. Their relationship was analyzed. Twenty healthy people were gathered as con- trols. Results The expression of miR155 was obviously elevated in peripheral blood and tissues of gastric adenoearcino- ma and it was increased along with clinical staging increasing. The expression of IGF-1 was obviously elevated in periph- eral blood and tissues of gastric adenocarcinoma and it was related with the invasion of gastric adenocarcinoma. And miR155 was related with IGF-1. Conclusion miR155 may participate in the pathogenesis of gastric adenocarcinoma and it may up-regulate IGF-1/IGF-1R signal transduction to promote the growth of gastric adenocarcinoma.
