胃癌组织中Keap1、IKKβ蛋白表达及意义

摘　　要：目的探讨Keich样环氧氯丙烷相关蛋白-1(Keap1)和IκB激酶β(inhibitor kappa B kinaseβ,IKKβ)在胃癌组织中的表达。方法应用免疫组化法检测54例胃癌患者的胃癌组织及24例癌旁正常组织中Keap1和IKKβ蛋白的表达,分析Keap1和IKKβ蛋白的表达与胃癌临床病理特征关系及二者表达的相关性。结果 Keap1与IKKβ在胃癌组织中的表达高于癌旁正常组织,差异具有统计学意义(P<0.05);胃癌组织中Keap1蛋白的表达与淋巴结的转移具有明显相关性(P<0.05),但与患者性别、年龄、肿瘤大小、组织学类型、组织分化程度无相关性(P>0.05);胃癌组中IKKβ蛋白的表达与组织分化程度及淋巴结转移相关(P<0.05),与患者性别、年龄、肿瘤大小、组织学类型均无相关性(P>0.05);Keap1的表达与IKKβ的表达无相关性(r=0.2839,P>0.05)。结论胃癌组织中Keap1、IKKβ蛋白表达异常,提示Keap1可能影响IKKβ介导的NF-κB信号通路,并在胃癌的发生、发展中发挥重要作用。Objective To investigate the expressions of Keapl and IKKβ in gastric carcinoma. Methods The. ex- pressions of Keapl and IKKβ were detected by immunohistochemistry in 48 cases of gastric carcinoma and 24 cases of adjacent normal tissue. The relationship of the expressions of Keapl and IKKβ with the clinicopathological features of gastric cancers was analyzed. Results The expressions of Keapl and IKKβ in gastric carcinoma tissue were higher than those in adjacent normal tissue （P 〈 0.05）. The expression of Keapl was significantly correlated with lymph node metastasis （P 〈 0.05） , but was not correlated with the gender, age, tumor size, histologic type, degree of differentia- tion （P 〉 0.05）. The expression of IKKβ was significantly correlated with lymph node metastasis and the degree of dif- ferentiation （P 〈 0.05） ,but was not correlated with gender, age, tumor size and histologic type. There was no correla- tion between the expressions of Keapl and IKKβ （r = 0. 2839, P 〉 0.05）. Conclusion The expressions of Keapl and IKKβ in gastric carcinoma are abnormal. The abnormal expressions of Keapl and IKKβ may induce the signaling path- ways of NF-κB, which result in the occurrence of gastric carcinoma.
