摘 要:目的探讨小檗碱对人T淋巴细胞白血病细胞株Molt-4细胞体外增殖抑制、凋亡诱导及相关机制。方法CCK-8法检测不同浓度的小檗碱对Molt-4细胞的增殖抑制作用;流式细胞术检测细胞凋亡率;RT-PCR检测Molt-4细胞Notch1、Bcl-xl、Deltex1基因表达情况。结果小檗碱诱导Molt-4细胞毒性及凋亡作用,呈剂量依赖性(P<0.05);RT-PCR显示Molt-4细胞株有Notch1 mRNA的表达,且随着小檗碱剂量的增加,Notch1、Bcl-xl、Deltex1 mRNA的表达逐渐下降。结论小檗碱可能通过Notch信号途径抑制Molt-4细胞增殖并诱导凋亡。Objective To investigate the effects of berberine on the proliferation,apoptosis and explore its mechanism in human T lymphoblastic leukemia cell line (Molt-4).Methods Molt-4 cells were treated with different concentrations of berberine in vitro respectively.Antiproliferative effect was detected by CCK-8 assay.Apoptosis rate was observed by flow cytometry (FCM).The expression of Notch1,Bcl-xl and Deltex1 mRNA was examined by Semi-quantitative reverse transcription PCR (RT-PCR).Results Berberine induced cytotoxicity and apoptosis in Molt-4 cells in a dose-dependent manner (P < 0.05).RT-PCR results showed that the expression of Notch1,Bcl-xl and Deltex1 genes decreased with the increase of berberine concentration (P < 0.05).Conclusion Berberine could inhibit the proliferation of Molt-4 cells and promote the apoptosis through down-regulation expression of apoptotic protein mediated by Notch signaling pathway.
