麦薏粉对荷瘤小鼠晚期的辅助作用及其机制

摘　　要：本文通过小鼠体内外实验,观察麦薏粉对荷瘤小鼠晚期的辅助作用.体外实验,采用MTT法测定麦薏粉对小鼠S-180瘤细胞抑制作用和巨噬细胞增殖作用,结果发现：麦薏粉对S-180瘤细胞抑制率达48.77%-87.60%,对巨噬细胞增殖作用介于5-氟尿嘧啶（5-FU）与盐酸左旋咪唑之间,最适宜用量为0.15 g/ml,利用该筛选出的适宜用量进行体内实验,建立小鼠S-180肿瘤晚期模型,用麦薏粉（0.15 g/ml）灌胃,连续10 d,并观察其对荷瘤小鼠瘤质量、肝脾质量及血细胞的影响,计算抑瘤率和肝脾指数,结果发现：麦薏粉可抑制肿瘤晚期小鼠体内肿瘤生长,抑瘤率达81.1%,可显著提高单核细胞、嗜酸性粒细胞的数量（P〈0.05）、降低嗜碱性粒细胞的数量（P〈0.05）,实验前后体质量差同正常组.本实验结果说明麦薏粉对肿瘤晚期小鼠有明显抑制肿瘤作用,平衡免疫功能,具有一定减少炎症反应及改善肿瘤晚期恶液质状况的作用.Through experiments of mice in vivo and in vitro,the auxiliary function of Wheat Adlay Powder on late stage tumor bearing mice was observed. MTT test was adopted to measure the inhibitory effect of Wheat Adlay Powder on S- 180 sarcoma cells and the proliferation promotion effect of Wheat Adlay Powder on Macrophages in vitro. Results showed that the inhibitory rate of Wheat Adlay powder on S- 180 cells on vitro was48. 77% ~ 87. 60%,and the macrophage effect was between 5- fluorouracil（ 5- FU） and levamisole hydrochloride,the suitable dosage for vivo experiment was 0. 15 g / ml. Mouse S- 180 late stage tumor model was built up with intragastric administration of Wheat Adlay powder（ 0. 15 g / ml） for 10 days continuously in vivo.By observing the influence of Wheat Adlay powder on the quality of tumor in tumor bearing mice,the quality of the liver spleen and blood cells,calculating the inhibition rate and the index of liver and spleen,it was revealed that Wheat Adlay powder could inhibit the tumor growth in vivo of the late stage mice of tumor,its inhibitory rate reached 81. 1%,and it could significantly improve the number of mononuclear cells cend,eosinophils（ P 0. 05）,and reduce the number of basophils（ P 0. 05）. Comparing to the results of experimental group,it didn＇t diverge very much before and after the experiment. The experimental results show that Wheat Adlay powder has an obvious inhibitory effect on the mice of late stage tumor and it can balance the function of immunity,make inflammatory reaction relieved to a certain extent,and improve the cachexia status on late stage cancer.
