摘  要:目的通过体外实验探讨过表达SEMA3G对人胰腺癌细胞PANC-1细胞增殖、侵袭和迁移能力的影响。方法将携带SEMA3G的慢病毒载体转染人胰腺癌PANC-1细胞系,利用real-time PCR和Western blot分别检测mRNA和蛋白质水平。CCK-8法检测细胞增殖,transwell实验检测细胞侵袭和细胞划痕试验检测细胞的迁移。结果成功建立稳定转染SEMA3G胰腺癌PANC-1细胞系。SEMA3G病毒感染组与空白对照组和阴性对照组相比细胞的增殖能力显著降低(P<0.05),SEMA3G病毒感染组的细胞侵袭和迁移能力明显低于两组对照组(P<0.05)。结论 SEMA3G慢病毒载体能有效过表达人胰腺癌PANC-1细胞中内源性SEMA3G蛋白,进而抑制细胞增殖、侵袭和迁移。Objective To investigate the effect of SEMA3G protein overexpression on proliferation, invasion and mi- gration of human pancreatic cancer PANC- 1 cells in vitro. Methods PANC- 1 cells of human pancreatic cancer were transfected with lentiviral vector carrying SEMA3G. The expression of SEMA3G at mRNA and protein level in cells was detected by real-time PCR and western blot, respectively. CCK-8 assay was applied to examine cell proliferation, the cell invasion was analyzed by Transwell assay and wound healing assay was used to examine cell migration in PANC-1 cells. Results The results showed that stable transfected SEMA3G cell line of pancreatic cancer line was successfully established. The proliferation capacity of SEMA3G group was significantly lower compared to blank and the negative control groups (P 〈0. 05). Similarly, cell invasion and migration capacity of SEMA3G group was signifi- cantly lower than the two kinds of control groups(P 〈0. 05). Conclusions SEMA3G lentiviral vectors can effectivelyincrease SEMA3 G protein expression in PANC-1 cells, thereby inhibiting cell proliferation, invasion and migration.
