摘  要:目的研究MiR200对肾癌细胞纤维化的影响机制,为鉴定治疗靶标提供理论依据。方法整群选取2013年1月—2015年1月该院收治的51例肾癌患者病例资料,于实验室,建立肾癌细胞株模型,采用癌旁正常组织作为对照,应用细胞侵袭实验和Westermblot法检测靶基因蛋白表达方法进行试验。结果 MiR-200组在SN12-PM6和786-0平均穿膜细胞的个数差异有统计学意义(P<0.05),MiR-200组ZEB1蛋白表达下降,而在E-cadberin蛋白表达上升,经相关系数比较,MiR-200的表达与E-cadberin成正相关(r=0.824)而ZEB1成负相关(r=-0.762)。结论 MiR-200能够调节皮脂纤维化的侵袭,降低ZEB1蛋白表达,从而E-cadberin蛋白表达上升。Objective To research the effect mechanism of MiR200 on renal carcinoma cell fibrosis and provide theoretical basis for identification and treatment of target. Methods The data of 51 cases of patients with renal carcinoma treated in the central laboratory of Qiqihar medical school were selected, the renal cell carcinoma strain model was built, the adjacent normal tissue was used for contrast, the target gene and protein expression was detected by cell invasiveness experiment and Westermblot method, and the correlation between the invasion ability of renal carcinoma cell of MiR-200 and ZEB1, E-cadberin and MiR-200 was analyzed. Results The difference in the average penetrating cell number of SN12-PM6 and786-0 in the MiR-200 group had statistical significance, P〈0.05, the ZEB1 protein expression in the MiR-200 group decreased, but the E-cadberin protein expression in the MiR-200 group increased, and the correlation coefficient comparison showed that the mi R-200 expression was positively correlated with E-cadberin,(r=0.824) but was negatively correlated with ZEB1,(r=-0.762). Conclusion MiR-200 can adjust the invasion of sebum fibrosis and reduce the ZEB1 protein expression thus increasing the E-cadberin protein expression.
