摘　　要：目的研究通用转录因子ⅡF多肽2(GTFⅡF2)的表达对食管癌细胞增殖及迁移的影响。方法人工合成靶向GTFⅡF2基因的siRNA序列和阴性对照siRNA序列及质粒pc DNA3.1-GTFⅡF2-FLAG,分别转染至食管癌细胞中,采用克隆形成实验和细胞划痕实验观察其对食管癌细胞增殖和迁移的影响。结果 RNA干扰技术靶向沉默GTFⅡF2基因表达后,食管癌细胞增殖和迁移均明显受到了抑制,而过表达GTFⅡF2对食管癌细胞的增殖和迁移无明显影响。结论GTFⅡF2基因在食管癌细胞TE-1、ECA-109的增殖和迁移过程中发挥重要作用,为进一步了解GTFⅡF2基因抑制TE-1、ECA-109细胞增殖和迁移的机制提供了基础。Objective To investigate the effect of general transcription factor GTFⅡF2 expression on the proliferation and migration of esophageal cancer cells. Methods Synthesized siRNA targeted to GTFⅡF2 gene or negative control siRNA and plasmid pc DNA3. 1-GTFⅡF2-FLAG were transfected to esophageal cancer cells respectively by using LipofectamineTM2000. Colony formation assay and wound-healing assay were performed to study the effect of knockdown and overexpression of GTFⅡF2 expression on the proliferation and migration of esophageal cancer cells respectively. Results Knockdown of GTFⅡF2 inhibited the proliferation and migration in TE-1,ECA-109 cells,but overexpression of GTFⅡF2 had no significant effect on cell proliferation and migration. Conclusion GTFⅡF2gene plays an important role in the proliferation and migration of TE-1,ECA-109 cells. It is useful for further understanding of the mechanism that GTFⅡF2 inhibits the proliferation and migration of TE-1,ECA-109 cells.
