摘 要:目的研究转移相关基因Notch1和VEGF在15例正常卵巢组织、32例卵巢良性肿瘤组织和48例卵巢恶性癌组织中的表达及相关性研究,旨在为卵巢癌的病理诊断及发病机制的探讨提供理论依据。方法采用免疫组化SP法检测各组Notch1、VEGF的表达情况和微血管密度(MVD),MVD采用CD34标记。并通过统计学方法分析其表达情况与临床病理特征的关系及相互间的关系。结果 Notch1在正常卵巢组织、卵巢良性肿瘤组织和卵巢恶性癌组织中的阳性表达率分别为13.33%(2/15)、31.25%(10/32)和81.25%(39/48);VEGF在正常卵巢组织、卵巢良性肿瘤组织和卵巢恶性癌组织中的阳性表达率分别为20%(3/15)、47.0%(15/32)和83.0%(40/48);CD34主要表达于各种卵巢组织的血管内皮细胞,MVD值在有淋巴结转移的卵巢癌组织中明显高于无淋巴结转移者(P<0.05)。通过Spearman相关性分析,Notch1和VEGF在卵巢恶性癌组织中呈现显著正相关(P<0.05)。结论 Notch1、VEGF是鉴别卵巢良恶性肿瘤的指标之一,检测淋巴结转移者中的MVD值对卵巢癌的恶性程度判断有帮助。Objective To investigate the expression and correlation of metastasis associated gene Notch1 and vascularendothelial growth factor(VEGF)in 15 normal ovarian tissues,32 samples of benign ovarian tumor,and 48 samples ofmalignant ovarian cancer in order to provide theoretical basis for pathological diagnosis and pathogenesis of ovarian cancer.Methods The expression levels of Notch1 and VEGF in each group were detected by immunohistochemical assay(SP),andmicrovessel density(MVD)was also detected with marker CD34. And the relationships of the expression levels with clinicalpathological features were analyzed by statistical methods. Results The positive expression rates of Notch1 were 13.33%(2/15),31.25%(10/32)and 81.25%(39/48)respectively,in the normal ovarian,benign tumor and malignant tumor tissues. Thepositive rates were 20%(3/15),47.0%(15/32)and 83.0%(40/48)separately in the above tissues. CD34 was mainly expressedin the vascular endothelial cells of various ovarian tissues MVD value was significantly higher in the ovarian cancer tissues withlymph node metastasis than those without(P〈0.05). Spearman correlation analysis showed that the expression levels of Notch1 and VEGF were positively correlated in ovarian cancer tissues(P〈0.05). Conclusion VEGF and Notch1 are the indicatorsfor the differentiation of benign and malignant ovarian tumors. It is helpful to detect the MVD value in malignancy evaluationfor ovarian cancer patients with lymph node metastasis.
