摘  要:目的研究紫薯花色苷提取物延缓果蝇衰老作用及其可能机制。方法以2 d龄Oregon K野生型雄性黑腹果蝇为模式生物,饲喂于含有不同浓度(0、0.5、1.0、2.0mg/ml)紫薯提取物的高脂培养基,定期更换新鲜培养基,并记录果蝇死亡数目。研究紫薯提取物对果蝇生存寿命,体内抗氧化酶活性[铜锌超氧化物歧化酶(copper-zinc-containing superoxide dismutase,SOD1);锰超氧化物歧化酶(manganese-containing superoxide dismutase,SOD2);过氧化氢酶(catalase,CAT)]、脂质过氧化产物丙二醛含量(malonaldehyde,MDA)、蛋白质羰基含量(protein carbonyl,PCO)及果蝇体内抗氧化相关基因表达水平的影响。结果与高脂模型组相比,紫薯提取物可以显著延长果蝇的平均寿命和最高寿命(2.0mg/ml,P<0.05);2.0mg/ml PSPE可以显著提高SOD1和CAT活力(P<0.05),极显著降低MDA和PCO含量(P<0.01);同时可以显著上调SOD1及转录因子NF-E2相关因子2(Nf-E2 related factor-2,Nrf2)m RNA表达水平(P<0.05)并且极显著上调SOD2和CAT m RNA表达水平(P<0.01),玛氏萨拉(methuselah,MTH)m RNA表达水平随PSPE浓度的增加呈下调趋势(P>0.05)。结论紫薯提取物能有效延缓机体衰老,同时对高脂膳食氧化损伤雄性果蝇具有一定保护作用,并可能通过改变体内相关抗氧活酶的活性及相关抗氧化基因表达水平来实现的。Objective To investigate the anti-aging effects of purple sweet potato extract (PSPE) and its possible protection mechanism in male high-fat Drosophila melanogaster. Methods Drosophila melanogaster aged 2d-old were fed different doses (0, 0.5, 1.0, 2.0 mg/ml) of PSPE in high-fat medium and then replaced with fresh medium regularly. The death rate was recorded. The lifespan, antioxidant enzymes activities, antioxidant encoding genes expressions and MDA as well as PCO content were detected. Results Compared with the model group, PSPE significantly extended the average lifespan and maximum lifespan of Drosophila melanogaster (2.0 mg/ml, P〈0.05); Treatment with 2.0 mg/mL PSPE significantly increased the Nrf2 and SOD1 mRNA expression levels (P〈0.05) as well as the SOD2 and CAT mRNA expression levels (P〈0.01). Enzyme activity assay showed that 2.0mg/mL PSPE significantly increased SOD1 and CAT antioxidant activity of PSPE was most likely associated with altering related antioxidant enzyme activity and regulating corresponding antioxidant gene expression.
