合格象皮及备选代用品成分分析

摘　　要：目的:找出与象皮成分相接近的代用品。方法:BCA法测定象皮和其它皮类样品中蛋白质含量,羟脯氨酸折算法测定象皮及其它候选代用品胶原蛋白含量,在HITACHI L-8800型氨基酸分析仪上检测样本的氨基酸组成与含量,检测方法依据GB/T 5009.124-2003。取象皮和其它代用品试样,粉碎成100目细粉,精确称取100mg,在Tecnai G2 F20透射电子显微镜上进行能谱分析。结果:象皮中所含金属元素(如镁、铝、钙等),较其它候选代用品高。各种候选代用品的元素组成与象皮相差较远。从水溶性总蛋白质含量分析,牛皮和驴皮与象皮含量接近。从总胶原蛋白含量分析,牛皮与象皮几乎相同。从氨基酸含量分析,在氨基酸总量及氨基酸组成上,从蛋白质和氨基酸指标总体分析,牛皮与象皮最为相近。结论:从蛋白质和氨基酸含量分析,在氨基酸总量、氨基酸组成以及蛋白质和氨基酸总体分析上,牛皮与象皮最为相近。Objective： To find out the substitutes whose components are similar to the elephant skin. Methods： Using BCA method to test the protein content contained in the elephant skin and other skins; according to GB/T 5009.124-2003, use HITACHI L-8800 analysis meter to test the components of amino acid of the specimen. To take the specimen of elephant skin and other substitutes, smash into 100 mesh, weight exactly 100 mg and then to make energy spectrum by Tecnai G2 F20 transmission electron microscope. Results： The metal elements, such as magnesium aluminum and calcium contained in the elephant skin is higher than those contaied in the substitutes. All the elements contained in the substitutes is far lower than those contained in the elephant skin.But under the following conditions, the cattle skin and the elephant skin are very similar, in water solubility protein contents. The cattle skin and the donkey skin is close to the elephant skin. Conclusion： In terms of the total collagen contents, the cattlle skin is very close to the elephant skin, in the components and the whole contents of the amino acid, the cattle skin is the closest to the elephant skin; in the general analyse of the index of the protein and the amino acid, they are rather close.
