摘  要:目的营养状态在癌症治疗中的作用一直受到广泛关注。该文旨在研究高营养状态对肿瘤细胞的影响。方法在肿瘤细胞的培养体系中添加不同浓度的胎牛血清(FBS,5%、10%、25%、50%),以模拟不同的营养状态。培养3周后,CCK-8法测定细胞增殖;克隆形成和体内成瘤实验评价肿瘤细胞成瘤性;最后通过油红染色、流式细胞术、电镜和蛋白质印迹检测探讨可能的机制。结果高血清浓度能促进人乳腺癌细胞系SKBR3细胞和人前列腺癌细胞系PC3M细胞的增殖,但会引起细胞空泡化,并抑制其克隆形成和成瘤能力。诱导后的PC3M细胞,自噬过程特征蛋白轻链3(LC3)Ⅱ/Ⅰ比例上调,电镜中观察到自噬小体增多,表明高浓度的血清可能通过激活自噬来抑制肿瘤细胞的成瘤性。结论高营养状态对肿瘤细胞成瘤性有明显抑制作用,有望成为新的癌症治疗策略。Objective To explore the effect of high nutritional status on tumor cells in response to the increasing attention on the role of nutritional status in cancer treatment. Methods Tumor cells were cultured in a medium supplemented with varied concentrations of fetal bovine serum( FBS) to simulate different nutritional status. Cell proliferation and tumorigenic capacity were assessed by Cell Counting Kit( CCK)-8,colony formation and xenograft tumorigenicity assay. Oil red O staining,flow cytometry,electron microscopy,and Western blotting analysis were used to investigate the possible mechanism.Results High FBS concentrations promoted the vacuolization of SKBR3 and PC3 M tumor cells,inhibited clonality and tumorigenicity but enhanced proliferation. Conversion of light chain 3( LC3) and observation of autophagic vacuoles in treated PC3 M cells indicated that excess nutrition was likely to play a specific role in inhibiting tumorigenicity through an autophagic pathway. Conclusion High nutritional status will potentially become a therapeutic strategy for cancers.
