摘  要:目的评价实时荧光PCR技术开展作为快速检测从业人员体检肠道致病菌(伤寒、痢疾)检测工作方法的适用性,以替代落后的细菌培养法检测手段的可行性。方法对一定时间从业人员体检的肛拭子,同时用PCR筛查法和传统培养法进行志贺菌和沙门菌对比检测,充分比较两者的检出率、灵敏度、特异性、工作效率、人力成本和耗材成本。结果 PCR筛查法阳性检出率1.6‰;传统培养法阳性检出率0.76‰;PCR筛查法的灵敏度和特异性分别为100%和99.97%,具有操作安全简便、结果客观性强、方法便于规范统一、质量控制措施可操作性强等特点。结论 PCR筛查法检测技术对健康体检人员的肠道致病菌进行快速筛查可以使检验的时间得以缩短,而且特异度强、灵敏度高,兼具时效性与客观性。更适合基层实验室规范统一地开展健康带菌检测工作。Objective Evaluation applicability of Real-time fluorescent PCR method to carry out a rapid detection from employees on physical examination of intestinal pathogenic bacteria (typhoid and dysentery). Methods Comparison test on anal swab of employees for a period of time by using the PCR method and the classic method at the same time . Fully com-pared their detection rate, sensitivity, specificity, efficiency, labor and material costs. Results The positive rate of PCR method is 1.6‰ and the classic method is 0.76‰. The sensitivity and the specificity of the PCR method is 100% and 99.97% respectively. Method is safe in operation to facilitate unified specification, quality control measures is strong oper-ability, etc. Conclusion Real-time fluorescent PCR method allows rapidly detection of intestinal pathogenic bacteria. The method is high specificity, high sensitivity. It is suitable for laboratory to carry out rapid detection from employees on physical examination of intestinal pathogenic bacteria normatively.
