摘 要:试验以禽白血病病毒感染汶上芦花鸡和正常汶上芦花鸡肝脏为研究对象,利用RNA-Seq技术对其进行高通量转录组测序,分析基因组结构信息及挖掘新转录本及新基因的功能,6个样品共获得254 413 928条有效数据(Clean Reads),总计32.05 Gb。得到可靠的SNP位点673 689个,其中位于基因区的SNP位点469 160个,基因间的SNP位点204 544个。6个样品中平均8 249个基因涉及可变剪接,其中外显子跳跃事件比例最大,其次为第一个外显子可变剪接事件,并对6 283个基因结构进行了优化,修正基因的边界。与所选鸡的参考基因组序列进行对比,共发掘新基因1 446个,其中1 058个新基因得到功能注释。研究进一步完善汶上芦花鸡的基因结构信息,为发掘潜在的新抗白血病基因提供分子水平依据。In the study,transcriptome of liver of Wenshang barred chicken infected with avian leukosis virus and healthy Wenshang barred chicken were sequenced using RNA-seq technology,and analyzed gene structure optimization, novel gene mining and function annotation. After quality control, 254 413 928 clean reads with a total of 32.05 Gb were obtained from the liver library. 673 689 SNPs were scanned, 469 160 SNPs of which were located in gene region and 204 544 were located in intergenic region. The average new alternative splicing of 6 samples were 8 249 genes. The gene structure of 6 283 genes was optimized by editing gene boundary. 1 446 novel genes were mined by comparing to the chicken reference genome. Function annotations of 1 058 novel genes were obtained by sequence alignment in different database. This study improved the gene structure information of Wenshang barred chicken and provided basis for mining novel anti-disease genes at molecular level.
