摘 要:目的利用双通道放电等离子体反应装置,研究常压空气等离子体对液相体系中金黄色葡萄球菌的杀灭规律和机制。方法观察等离子体放电参数对金黄色葡萄球菌杀灭的剂量效应。从化学(羟基自由基)、物理(水体温度、紫外光)和菌体微观形态(透射电镜)等角度综合分析等离子体场中各活性物质的杀灭作用。结果常压空气等离子体液相杀菌效果与放电时间呈正相关关系。当电极距液面间距为1 mm,放电处理10 min时,杀灭率达99.91%。透射电镜技术观察金黄色葡萄球菌菌体形态和超微结构,发现杀菌后菌体变得相对模糊,出现明显变形、破裂甚至穿孔现象,细胞质流向周围介质。杀菌过程中羟基自由基起主要作用,而紫外光杀菌效应及热力杀菌效应仅起协同作用。结论该装置能实现对液相环境金黄色葡萄球菌杀灭,在等离子体场的强烈作用下细菌细胞发生破损是造成细菌死亡的主要原因。Objective To observe the disinfection effect of Staphylococcus aureus in liquid exposed to atmospheric pressure dual channels discharge air plasma.Methods The germicidal effect(GE) of Staphylococcus aureus in water in the variation range of discharge parameters by atmospheric pressure air plasma was investigated.Temperature,UV,·OH and bacterial cells cracking were tested to reveal the roles of active radicals.Results There was a positive correlation between the GE and treatment time.When The distance between electrode and liquid was 1 mm with treatment time of 10 min,the GE value could reach 99.91%.Results of TEM showed that the cell wall(cell membrane) cracked,cell permeability changed and cell content leaked.The disinfection mechanism of atmospheric pressure air plasma in liquid mainly dependsed on ·OH created in discharging,while the heat and UV radiation played limited roles.Conclusion Atmospheric pressure air plasma can kill Staphylococcus aureus in liquid efficiently in short time,and cracking of bacterial cells is the main cause of cell death.
