摘 要:[目的]观察参七消痞颗粒对慢性萎缩性胃炎(CAG)大鼠COX-2、Bcl-2蛋白表达的影响。[方法]通过N-甲基-N-硝基-N-亚硝基胍(MNNG)负荷其他因素建立CAG大鼠模型(14周),之后再随机分为模型组、摩罗丹组(阳性对照组1)、叶酸组(阳性对照组2)、参七消痞颗粒高、中、低剂量组,另从实验开始造模时就设立正常对照组(空白组)。各组大鼠给予相应药物灌胃治疗8周。治疗结束后,苏木精-伊红染色观察胃黏膜病理改变,ELISA法大鼠血清中PGI、PGII水平,并计算PGI/PGII;使用放射免疫法检测大鼠血清中GAS水平,免疫组织化学法检测大鼠胃组织COX-2、Bcl-2蛋白表达。[结果]与模型组比较,参七消痞颗粒中、低剂量组与模型组比较血清GAS水平显著降低(P<0.01、P<0.05),与空白组比较,模型组大鼠胃组织COX-2、Bcl-2蛋白表达均明显升高(P<0.01),而参七消痞颗粒各剂量组及阳性对照组均较模型组降低(P<0.01、P<0.05)。[结论]参七消痞颗粒能明显改善CAG大鼠的胃黏膜病变,其治疗机制可能与下调COX-2、Bcl-2蛋白的表达有关。[Objective]to evaluate the effect of Shenqi Xiaopi(SQ)granules on the expression of COX-2and Bcl-2in rats with chronic atrophic gastritis.[Methods]CAG model was established by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG)and other comprehensive factors for 14 weeks.The model rats were randomly divided into 6groups,namely the model group,the Moluodan group,the folic acid group and the SQ granules 2,4,8g/kg groups.At the same time,the normal wistar rats were taken as control group.After eight weeks' intragastric administration,gastric mucosa sample were taken for hematoxylin-eosin(HE)staining.Serum PGⅠ,PGⅡlevels were detected by Elisa assay and GAS was measured by radioimmunoassay.The protein expression levels of COX-2and Bcl-2were examined by immunological histological chemistry assay.[Results]Compared with the model group,serum levels of GAS in the SQ Granules 2g/kg and4g/kg groups were significantly decreased(P〈0.01、P〈0.05),the expression of COX-2and Bcl-2were significantly increased in the model group(P〈0.01)and they were down-regulated in all the treatment groups(P〈0.01、P〈0.05).[Conclusion]SQ granules can obviously improve mucosal atrophy in rats with atrophic gastritis,the mechanism is possibly associated with down-regulated the expression of COX-2and Bcl-2.
