摘  要:目的研究茶多酚(TP)对心脏骤停(CA)后心肺复苏(CPR)大鼠脑神经细胞凋亡的影响。方法健康SD大鼠随机分为假手术(SH)组、生理盐水(NS)组、茶多酚(TP)组,除SH组外,其他组均经食道交流电刺激诱发心室颤动,5min后行CPR,待自主循环恢复(ROSC)后即刻给予TP或生理盐水,ROSC后12、24、48、72h取各组大鼠的皮层和海马。分别检测各时间点原位末端转移酶标记技术(TdT—mediated dUTP nick end labeling)和caspase3荧光密度及caspase12的Westernblotting表达。结果TP组和NS组72h时TUNEL和caspase3荧光密度均大于SH组(P〈0.05),而TP组Tunnel和caspase3荧光密度均小于Ns组(P〈0.05)。与Ns组和SH组比较,TP组48、72hcaspase12表达明显减少(P〈0.05),NS组的表达高于SH组(P〈0.05)。结论TP可抑制心肺复苏后缺血再灌注引起的神经细胞凋亡,与其降低caspase12和caspase3的激活有关。Objective To study the effect of tea polyphenols (TP) on nerve cell apoptosis in rats with cardiac arrest following cardio - pulmonary resuscitation (CPR). Methods Healthy SD rats were randomly divided into sham (SH)group, normal saline (NS) group, and tea polyphenols (TP) group. In addition to the SH group, others were induced ventricular fibrillation esophagus stimulation with alternating current. Five min after cardiac arrest CPR was performed. Once restoration of spontaneous circulation (ROSC) was gained, TP or NS were immediately administrated. Cortex and hippocampus of rats in each group were collected at 12, 24, 48, 72 h after ROSC. TUNEL( TdT mediated dUTP Nick end labeling) and caspase 3 fluorescence intensity, as well as caspase 12 expression of Western blotting at each time point were measured respectively. Results TUNEL and caspase 3 fluorescence density at 72 h of TP group and NS group were greater than SH group (P 〈 0.05 ), while the TUNEL and caspase 3 fluorescence density of TP group were smaller than that of NS group ( P 〈 0. 05 ). Caspase 12 expression of TP at 48, 72 h decreased significantly when compared with NS group and SH group ( P 〈 0.05), and the expression of NS group was higher than that in SH group (P 〈 0.05). Conclusion TP can inhibit nerve cell apoptosis caused by ischemia -reperfusion after CPR, which relates to decreasing the activation of caspase 3 and caspase 12.
