毛霉菌丝提取物检测特性的研究

摘  要:试验主要为解决HPLC测定毛霉菌丝提取物的提取溶剂选择和检测器选择的问题,采用8种溶剂对毛霉菌丝进行提取,提取后分别用紫外分光光度计、荧光分光光度计和折光仪进行检测,通过数据分析得出结果,再上HPLC进行验证检测。结果表明:所有溶剂提取物的折光指数与空白和纯溶剂相比变化不大,不适宜在HPLC检测时采用示差折光检测器检测。而紫外检测特性和荧光检测特性的分析表明:50%的甲醇溶液、50%的乙腈溶液和AMF这3种溶剂提取物有较好的信号响应,进一步进行这3种溶剂毛霉菌丝提取物的HPLC上机试验。结论是可选用50%的甲醇溶液作为提取溶剂,在配备紫外检测器的高效液相色谱仪上进行检测,检测波长设置为215.5nm。The main purpose of this experiment is to select the extraction solvent and the detector for HPLC detection of Mucor mycelium extract. Eight kinds of solvent is used to extract Mucor mycelium and the extracts are determined by UV spectrophotometer, fluorescence spectrophotometer and Ahbe refractometer respectively. Some extracts with good detection characteristics are verified by HPLC. The results show that the refractive index of all eight extracts show little difference with blank and pure solvent, which indicates that refractive index detector is not suitable for HPLC detection of Mucor mycelium extract. Extracts of 50% methanol solution, 50% acetonitrile and AMF have good UV and fluorescence signal response. Further tests of the three solvent extracts are processed on HPLC, and the conclusion is that Mucor mycelium extracted by 50% methanol solution and detected by UV detector at 215.5 nm would be well.
