摘  要:目的分析不同脱钙条件对骨组织免疫组织化学(免疫组化)染色抗原性的影响。方法用8%硝酸分别于微波、光波及光波+微波组合Ⅰ、光波+微波组合Ⅱ的条件下对骨组织进行脱钙,并将室温(20℃)条件下脱钙的骨组织作为对照组,运用链霉菌素卵白素-生物素(LSAB)免疫组化染色技术,并对其染色效果进行分析比较。结果微波、光波、光波+微波组合Ⅰ及光波+微波组合Ⅱ的脱钙时间显著低于对照组,且免疫组化染色效果显著优于对照组(P<0.05);光波+微波组合的免疫组化染色效果显著优于微波、光波(P<0.05);但微波与光波之间、光波+微波组合Ⅰ与光波+微波组合Ⅱ之间的免疫组化染色效果比较,差异无统计学意义(P>0.05)。结论光波+微波组合脱钙时间要明显短于微波、光波及室温脱钙,且具有良好的免疫组化染色效果。ObjectiveTo analyze influence by different decalcification conditions on bone tissue antigenicity in immunohistochemical staining.MethodsDecalcification was made through 8% nitric acid under microwave, light wave, light wave + microwave Ⅰ and light wave + microwave Ⅱ in bone tissue. Decalcification under room temperature (20℃) was taken as control group. Labelled streptavidin biotin method (LSAB) immunohistochemical staining was applied to analyze and compare stain effects.ResultsDecalcification time under microwave, light wave, light wave + microwave Ⅰ and light wave + microwave Ⅱ were all shorter than control group, and their immunohistochemical staining effects were all better than control group (P〈0.05). Light wave + microwave had better immunohistochemical staining effects than single microwave or light wave (P〈0.05). There was no statistically significant difference of immunohistochemical staining effects across microwave, light wave, light wave + microwave Ⅰ and light wave + microwave Ⅱ (P〉0.05).ConclusionLight wave + microwave provides shorter decalcification time than microwave, light wave and room temperature, and it contains good immunohistochemical staining effect.
