摘  要:分析细胞质遗传背景是马铃薯育种亲本选配和早世代选择工作的重点。以4个携带不同细胞质遗传背景的马铃薯栽培品种为实验材料,利用已开发的cpDNA/mtDNA多态性标记,建立了在一个PCR反应体系中能同时鉴别不同细胞质遗传背景的多重PCR扩增体系。结果表明:多重PCR和单一PCR扩增体系对马铃薯cpDNA上4个位点(ndh C/trn V、rps16/trn Q、1/11a、10)和mtDNA上位点Band 1的扩增及经Bam H I酶切后的片段一致。用该体系鉴别大规模群体的细胞质遗传背景,可大大降低实验成本,节约时间、人力和物力,在马铃薯种质资源鉴定和分子标记辅助选育中具有重要的应用价值。Detecting cytoplasmic background was one of the most important works in potato breeding program. Using four potato varieties with different cytoplasmic types as materials and five specific primers reported for cytoplasmic background detection,the author developed a multiplex PCR system which could detect different cytoplasmic background of potatoes simultaneously. The results showed that the amplified products and the digested products by Bam H I of four loci( ndh C / trn V,rps16 / trn Q,1 /11 a,10) on cpDNA and Band 1 on mtDNA of tested potato varieties were accordant by the multiplex and single PCR system. This multiplex PCR system would be very important for germplasm identification and for marker-assisted selection during early stage of potato breeding,because of time saving,less labor and lower cost for the experiment. It thus could be a significant method of potato breeding program.
