黑面神不同极性部位HPLC色谱与抗慢性皮炎-湿疹疗效的关联性研究

摘　　要：目的:研究黑面神不同极性部位高效液相色谱与其抗慢性皮炎-湿疹疗效的关联性,为探索黑面神的有效物质基础提供参考依据。方法:采用高效液相色谱法对黑面神不同极性部位进行分离,采用梯度洗脱程序,流速为1.0mL·min^(-1),柱温为30℃,进样量为20μL。建立慢性皮炎-湿疹小鼠模型,观察黑面神不同极性部位对小鼠耳肿胀的改善作用。结果:黑面神乙酸乙酯萃取部位和正丁醇萃取部位所含成分的种类和数量显著多于氯仿部位和剩余水层部位,且这两个部位抗慢性皮炎-湿疹疗效亦优于氯仿部位及剩余水层部位,差异均具有统计学意义(P<0.05或0.01)。结论:黑面神抗慢性皮炎-湿疹作用的效应物质主要来源于其乙酸乙酯和正丁醇萃取部位,其高效液相色谱及相关峰数据可用于衡量黑面神的抗慢性皮炎-湿疹疗效。Objective： Correlation study on HPLC and therapeutical effect of dermatitis-eczema of different polar parts of Breynia Fruticosa in order to further research the effective material bases for the treatment of chronic dermatitis-eczema. Methods：Using HPLC to analyze the different polar parts of branches and leaves of Breynia Fruticosa. Chromatographic conditions： gradient program, flow rate：l. 0mL · min-1 ; temperature 30℃, and the injection volume..20btL. Observe the therapeutic effect of mice ear swelling by different polar parts of branches and leaves of Breynia Fruticosa. Results： The amount and type of chemical compound in the ethyl acetate and n-butyl alcohol part were more significant than those in the Chloroform part and the rest of the water area. These two polar parts had a better therapeutic effect. Conclusion： The active constituents of the treatment of chronic dermatitis eczema are mainly from the ethyl acetate and n-bu- tyl alcohol part, and it can be used to judge the therapeutic effects of chronic dermatitis eczema.
