摘 要:目的研究补阳还五汤(BYHWD)对失神经肌萎缩SD大鼠神经生长因子(NGF)分泌的影响。方法选取雄性SPF级SD大鼠48只,随机分为6组,补阳还五汤高中低剂量组、模型组、假手术组,弥可保组。分别建立腓总神经夹伤模型,手术后每日灌胃,21d后测量足趾宽度,记录展指功能;进行肌电图检测,测量肌电位衰减率;检测胫前肌的湿重比;运用免疫组化法检测大鼠NGF的分泌情况。结果 1展趾功能测定:BYHWD高剂量组、弥可保组与模型组比,显著增高(P<0.01或P<0.05);2电位衰减率:BYHWD高剂量、弥可保组与模型组比较差异显著(P<0.0001或P<0.01)。中药高剂量与中、低剂量组相比较也具备了极显著差异(P<0.0001或P<0.01);3胫前肌湿重比:BYHWD高剂量、弥可保组与模型组相比,均有显著增高(P<0.05);4 NGF蛋白的分泌:假手术组、中药各组和弥可保组可见较多NGF免疫反应阳性颗粒,呈浅棕色。模型组大鼠的NGF阳性颗粒数目减少,着色浅淡。BYHWD高中低剂量组、弥可保组与模型组比差异显著(P<0.01或P<0.05)。结论补阳还五汤能明显降低胫前肌湿重比的退变和电位衰减率,促进展趾功能的恢复,对失神经肌萎缩有很好的治疗效果,其机制可能是通过促进NGF蛋白的分泌实现的。Objective To explore the influence of Buyanghuanwu decoction( BYHWD) secretion of nerve growth factor( NGF)on the denervated muscle atrophy in rats. Methods 48 male Sprague- Dawley rats were subjected to common peroneal nerve crush model,then they were randomly divided into sham operation group,model group,BYHWD high- dose,medium- dose,low- dose groups,mecobalamin group( positive control). Each group has eight rats. The gavage lasted for 21 d. After that,toe width was measured,recording exhibition refers to the function; Then EMG testing were performed,measuring RNS decay rate; The wet weight ratio of tibial muscle was measured; At last,immunohistochemical staining secretion in rat NGF protein were measured.Results 1Exhibition toe function test: compared with model group,BYHWD high- dose group,mecobalamin group was significantly increased( P 〈0. 01 or P〈 0. 05) 2RNS decay rate: compared with model group,BYHWD high- dose group,mecobalamin group performed significant difference( P 〈0. 0001 or P〈 0. 01). Between high- dose and low- dose group,there existed important difference( P 〈0. 0001 or P 〈0. 01) 3 Wet weight ratio: compared with the model group,BYHWD high- dose,mecobalamin group were increased obviously( P〈 0. 05) 4 NGF secretion: more NGF immunoreactive particles were seen in sham group,BYHWD groups mecobalamin group,coloring brown. The model group NGF reduced the number of positive particles,coloring pale. In the comparison with the model group,BYHWD doses and mecobalamin groups made statistically significant( P〈 0. 01 or P 〈0. 05). Conclusion BYHWD can effectively delay denervated tibial muscle atrophy of rats by increasing the wet weight ratio,toe function and defering the RNS delay rate which mechanism maybe increase the promotion of NGF protein secretion.
