摘  要:目的探讨丹红注射液对血管性痴呆(VD)大鼠行为学及海马CA1区神经细胞凋亡的影响。方法筛选造模成功的VD大鼠随机分为治疗组和VD模型组,每组12只大鼠,另有12只正常组、12只假手术组。治疗组给予丹红注射液腹腔注射给药;同时正常组、假手术组、VD模型组分别腹腔注射生理盐水。28d治疗结束后各组大鼠开始做行为学测试,行为学测试结束后,行免疫组织化学检测Bcl-2、Bax阳性细胞的表达。结果与VD模型组相比,治疗组大鼠逃避潜伏期明显缩短,穿越目标象限时间明显延长,差异有统计学意义(P<0.05);治疗组与VD模型组大鼠海马区Bcl-2和Bax蛋白表达显著高于假手术组与正常组,而治疗组与VD模型组相比,Bcl-2蛋白表达增加的幅度明显大于Bax蛋白(P<0.05)。结论丹红注射液通过调节Bcl-2、Bax蛋白表达来抑制细胞凋亡,从而改善VD大鼠的学习以及认知能力。Objective To study the effect of Danhong injection on rats behavior with vascular dementia and ap- optosis of neuronal cells in the hippocampal CA1. Methods Successful model of rats VD were randomly divided in- to a treatment group and a VD model group. The treatment group received Danhong injection,while the normal group,sham-operation group,and VD model group were injected with saline. 28 days later after the treatment be- havioral tests were performed among these rats, and then the expressions of Bcl-2 and bax were detected by immu- nohistochemistry. Results Compared with the VD model group,the escape latent period was significantly shorter in the treatment group,and the crossing target quadrant time was prolonged (P〈0.05). The expressions of Bcl-2 and Bax protein in the hippocampus of the VD model group and the treatment group were significantly higher than those in the sham-operation group and the normal group. Compared with the VD model group,the increase of Bcl- 2 expression in the treatment group was significantly greater than the increase of Bax (P〈0.05). Conclusion Danhong injection can improve the ability of learning and cognizance by regulating the expressions of Bcl-2 and Bax to inhibit apoptosis of neuronal cells.
