摘  要:转录因子Tbx18在胚胎心脏发育过程中起重要调控作用,是心外膜祖细胞标记之一;故以Tbx18为标记的阳性祖细胞群被称为:Tbx18+心外膜祖细胞(epicardial progenitor cells,EPCs)。小鼠胚胎、新生和成年期心脏组织细胞的特性区别较大,成年小鼠的心脏属于终末分化组织。但是,Tbx18+EPCs对成年小鼠心脏组织的贡献大小尚存争议。本研究拟定量分析Tbx18+EPCs对成年小鼠心脏组织的贡献大小。采用整体和组织切片X-gal染色检测成年心脏组织Lac Z的表达;荧光激活细胞分选法(fluorescence activated cell sorting,FACS)分离成年Tbx18Cre/R26EYFP小鼠心脏组织EYFP+细胞。结果显示,在Tbx18+EPCs遗传谱系示踪小鼠,报告基因Lac Z和EYFP在成年小鼠心脏的心室、心房、冠状动脉、室间隔等处表达;成年Tbx18Cre/R26EYFP小鼠心脏组织细胞用FACS分离,分选的EYFP+细胞比例平均约为33.94%。由此可见,成年小鼠心脏的心室、心房、冠状动脉、室间隔等心脏组织均可来源于Tbx18+EPCs;约1/3成年小鼠心脏组织细胞来源于Tbx18+EPCs。故Tbx18+EPCs参与成年小鼠心脏组织的部分形成。Transcription factor Tbx18 plays a vital role in controlling the development of embryonic heart,and it is one of the markers of epicardial progenitor cells;therefore,those positive progenitor cell groups marked with Tbx18 are named as "Tbx18+EPCs(epicardial progenitor cells) ".The embryonic,new born and adult mice's cardiac histocytes differed a lot in their characteristics,and the heart of an adult mouse is already ultimate differentiated tissue.However,researchers are still debating about the role of Tbx18+EPCs in mice's cardiac development.In this study,we tried to do a quantitative analysis of its role in adult mice's cardiac development.We tried to examine the Lac Z expression in adult cardiac tissue through observing whole mount and tissue sections ' X-gal staining method,and by using fluorescence activated cell sorting(FACS) method to sort out Tbx18 Cre/R26EYFPmouse's EYFP+cells in heart.The results showed that on Tbx18+EPCs-genetic-lineage-tracing-mouse,reporter gene EYFP and Lac Z were expressed in ventricles,atria,coronary artery,interventricular septum in their adults' heart;the cardiac cells are isolated by FACS in adult Tbx18 Cre/R26 EYFPmouse,the positivity ratio of EYFP+cells is 33.94% in average.Thus,the histiocyte of adult mouse's ventricle,atrium,coronary artery,interventricular septum can be derived from Tbx18+EPCs;about 1/3 of adult mouse's cardiac histocytes may originate from Tbx18+EPCs.Therefore Tbx18+EPCs partially involved in the formation of the cardiac tissue of adult mice.
