胃癌组织中a2V表达及与M2型巨噬细胞的相关性

摘　　要：目的探讨液泡膜-ATP酶a2亚基（a2V）在胃癌组织中的表达,及其与M2型巨噬细胞水平的相关性,并探讨a2V表达与胃癌生物学行为的关系。方法应用免疫组化方法检测胃癌组织和癌旁组织中a2V的表达,以CD163标记M2型巨噬细胞,检测其在胃癌组织中的水平,结合病人的临床病理资料分析两者的相关性及a2V与胃癌生物学行为的关系。结果胃癌组织中a2V的阳性表达率高于对应的癌旁正常组织（χ~2=9.751,P〈0.05）。胃癌组织中a2V表达与CD163水平呈正相关（r=0.360,P〈0.05）。a2V的表达与胃癌组织TNM分期、淋巴结转移有关（χ~2=7.581、6.289,P〈0.05）。a2V低表达胃癌病人的生存情况优于高表达病人（χ~2=4.604,P〈0.05）。结论 a2V在胃癌组织中表达上调,与胃癌的侵袭和转移及M2型巨噬细胞的水平有关,a2V蛋白高表达病人的预后不良。Objective To investigate the correlation between the expression of a2V and M2-macrophages in gastric can-cer（ GC）,and explore the relationship between a2V expression and biological behavior of this malignancy. Methods Immunohistochemical method was used to detect the expressions of a2V in GC and its paraneoplastic tissue,the M2-macrophages were labeled with CD163,and its level in GC was determined. The correlation between a2V and M2,as well as the association of a2V with biological behaviour of GC were analyzed combining with patients’ clinicopathological data. Results The positive rate of a2V expression in the cancer tissue was higher than that in the corresponding paraneoplastic normal tissue（ χ~2= 9. 751,P 〈 0. 05）. In the GC tissue,a2V was positively correlated with CD163（ r = 0. 360,P 〈 0. 05）. The a2V expression was associated with TNM stage of GC and its lymph node metastasis（ χ~2= 7. 581,6. 289; P 〈0. 05）. Patients with low a2V expression had a higher survival rate than those with high a2V expression（ χ~2= 4. 604,P 〈 0. 05）. Conclusion The expression of a2V is up regulated in gastric cancer,which is associated with the invasion and metastasis as well as the level of M2-macrophages. Patients with high expression of a2V have a poor prognosis.
