Hippuristanol对卵巢癌细胞生物学特性的影响

摘　　要：目的探讨真核细胞起始因子e IF-4A抑制剂Hippuristanol(Hipp)体外对人卵巢癌细胞增殖、侵袭及凋亡的影响。方法将人卵巢癌A2780细胞体外培养,采用MTT法检测不同浓度(0、25、50、100、200 nmol/L)Hipp不同时间(24、48、72 h)作用下A2780细胞增殖情况的变化,采用Transwell小室侵袭实验及流式细胞仪检测不同浓度(0、25、200 nmol/L)Hipp作用72 h后卵巢癌细胞侵袭及凋亡情况的变化。结果与空白对照组相比,不同浓度及不同时间作用下的Hipp均能抑制A2780细胞增殖,且呈时间和剂量依赖性;当主效应为浓度时,随着Hipp浓度的增加,A2780细胞的增殖抑制率明显升高(F=10 305.97,P<0.05);当主效应为时间时,随着Hipp作用时间的延长,A2780细胞的增殖抑制率也明显升高(F=1 747.85,P<0.05)。Hipp能使G1期细胞比例增加,S、G2/M期细胞比例减少,且随浓度增加效果更加明显,差异有显著性(P<0.05);细胞凋亡率亦随Hipp浓度增加而增大,各组间比较差异有显著性(P<0.05);透膜细胞数随Hipp浓度增加明显减少,各组间比较差异有显著性(P<0.05)。结论Hipp能显著抑制卵巢癌细胞A2780的增殖、侵袭并加速其凋亡,Hipp有望成为临床靶向治疗卵巢癌的新药物。Objective To investigate the effects of Hippuristanol（ Hipp） on cell multiplication,invasion and apoptosis of human ovarian cancer in vitro. Methods Human ovarian cancer A2780 cells were cultured in vitro. Employing MTT method,the proliferation of A2780 cells under different concentrations（ 0,25,50,100,200 nmol / L） of Hipp and different time（ 24,48,72 h） were detected.Applying Flow cytometry and Transwell,the invasion and apoptosis of A2780 cells were detected under the action of different concentrations（ 0,25 and 200 nmol/L） of Hipp for 72 h. Results Compared with the blank control group,Hipp could inhibit the proliferation of A2780 cells at different concentrations and time,showing time-dose dependent. When the main effect was concentration,the inhibitory rate of A2780 cell multiplication increased along with the increase of Hipp concentration（ F = 10 305. 97,P 〈 0. 05）; when the main effect was time,the inhibitory rate of A2780 cell multiplication also increased along with the extension of time（ F = 1 747. 85,P 〈 0. 05）. Hipp increased the proportion of satge-G1 cells,and decreased the proportion of stage-S and stage-G2/ M cells,the effectiveness was more significant with the increase of Hipp concentration,and the apoptosis of the cells also increased with the increase of Hipp concentration. The number of permeable membrane cells decreased when the concentration of Hipp increased,the difference among each group was significant（ P 〈 0.05）. Conclusion Hippuristanol can significantly inhibit the proliferation and invasion of A2780 cells of ovarian cancer and accelerate the apoptosis of the cells,which is expected to become a new drug for targeted therapy of this malignancy.
